川芎嗪对宫内生长受限幼鼠学习记忆能力的影响

摘    要:目的:探讨川芎嗪对宫内生长受限(FGR)幼鼠学习记忆能力的影响。方法:被动吸烟法建立大鼠FGR模型。孕鼠随机分为5组:对照组、模型组、川芎嗪高、中、低剂量治疗组。记录幼鼠出生体重;于23日龄开始Mor-ris水迷宫试验,29日龄开始消极回避试验,测试其学习记忆能力,30日消极回避试验后测量幼鼠体重。结果:①模型组幼鼠出生体重低于对照组及各川芎嗪治疗组,差异有显著性(P<0.01)。②Morris水迷宫试验中,训练第2-6天,模型组幼鼠平台潜伏期均高于对照组及川芎嗪各剂量组;消极回避试验中,模型组幼鼠所受电击次数高于对照组及川芎嗪各剂量组(低剂量组P<0.05;中、高剂量组P<0.01)。模型组24 h后保持潜伏期低于对照组及川芎嗪各剂量组(低剂量组P<0.05;中、高剂量组P<0.01)。③30 d体重模型组明显低于对照组及川芎嗪各剂量组(低剂量组P<0.05;中、高剂量组P<0.01)。结论:FGR幼鼠存在生长发育障碍及学习记忆能力下降,川芎嗪可促进幼鼠生长发育及学习记忆能力。

Abstract:Objective: To explore the effect of tetramethylpyrazine on learning and memory in rats of fetal growth restriction(FGR).Methods: The model of pregnant rat with FGR was established by passive smoking method.Forty pregnant rats were randomly divided into five groups: control group,FGR group, high-,middle-,and low-dose of tetramethylpyrazine treatment group.The body weight of rats was measured on postnatal day(PD) 1.By Morris water maze test from PD23 to PD28 and passive avoidance procedures on PD29,and the learning and memory ability of young rats was detected.The body weight was measured on PD30.Results: ① The body weight on PD1 of the FGR was lower than that of the control group and the tetramethylpyrazine treated groups(P<0.01).② Compared with the control group and tetramethylpyrazine groups,the platform latency of the FGR group increased from training day 2 to 6.The number of foot shocks of the FGR group was higher than that of the control group and tetramethylpyrazine groups((P<0.05),versus low-dose of tetramethylpyrazine group;and both P<0.01,versus middle- and high-dose of tetramethylpyrazine group),while the retention latency of the FGR was lower than that of the control group and tetramethylpyrazine groups.③ The body weight on PD30 of the FGR was lower than that of the control group and the tetramethylpyrazine groups.Conclusion: FGR rats presented developmental disorders and lower learning and memory ability.Tetramethylpyrazien can improve the development,learning and memory of young rats.
