Smads蛋白家族在慢性哮喘大鼠气道重建中的表达及中药麻芩汤干预作用的研究

摘    要:目的:探讨Smads蛋白家族在慢性哮喘大鼠肺组织中蛋白和mRNA变化情况及中药复方麻芩汤的干预作用。方法:实验设正常组、模型组和中药组,各组予相应处理2周;RT-PCR测定大鼠肺组织Smads家族mRNA的变化情况;WesternBlotting方法测定大鼠肺组织Smads蛋白含量的变化情况。结果:RT-PCR测定结果显示:模型组Smad2mRNA较正常组高表达(P<0.05),中药组较正常组高表达(P<0.05);模型组Smad7mRNA较正常组低表达(P<0.05),而中药组与正常组相比无显著差异;Smad4mRNA表达3组之间无差异(P>0.05)。WesternBlotting方法检测结果显示:模型组Smad2较正常组显著高表达(P<0.05),模型组Smad4、Smad7较正常组显著低表达(P<0.05);中药组Smad2较正常组显著高表达(P<0.05),Smad4、Smad7较正常组无显著差异。结论:在慢性哮喘发生中,Smads蛋白家族的Smad2、7均发生显著变化,中药麻芩汤对此有调节作用。

EXPRESSIONS OF SMADS FAMILY PROTEIN IN AIRWAY REMODELING OF CHRONIC BRONCHIAL ASTHMA RATS AND INTERVENE EFFECT OF MAQIN DECOCTION

Abstract:Objective: To explore the effect of TGF - α /Smad signal pathway in airway remodeling of chronic bronchial asthma rats by observing the changes of Smads protein family expressions in lung tissue of rats with chronic bronchial asthma. Methods: 24 SD rats were divided into three groups,normal,model and Chinese medicine groups,models were built by injecting 10% ovalbumin and 10% aluminum hydroxide,normal and model group were given 0. 9% saline,Chinese medicine group was given Maqin decoction for 2 weeks. the changes of Smads mRNA in lung tissue was determined by RT - PCR,the content of Smads protein in lung tissue was detected by Western Blotting. Results: RT - PCR examination results showed that the expression of Smad 2mRNA in lung tissue of rats with chronic bronchial asth- ma was higher than that of normal,that expression in Chinese medicine group was higher,too; the expression of Smad 7mRNA in lung tissue of rats with chronic bronchial asthma was obviously lower than that of normal,there was no difference between the normal and Chinese medicine groups on the expressions of Smad 7mRNA; the expression of Smad 4 mRNA had no obvious changes in the three groups. Western Blotting results showed that the expression of Smad 2 protein in lung tissue of rats with chronic bronchial asthma was higher than that of normal,the expressions of Smad 4 and Smad 7 protein were lower than those in normal; the expression of Smad 2 protein in lung tissue of Chinese medicine group was higher than that of normal,there were no difference between Chinese medicine group and normal group on the expressions of Smad 4 and Smad 7 protein. Conclusion: Smad 2 and Smad 7 of Smads family changed obviously in the lung tissue of rats with chronic bronchial asthma,which were closely relate to the airway reconstruction,Chinese medicine can regulate the changes.
