胃癌组织中VEGF和CD34的表达及微血管密度的临床病理意义

目的探讨血管内皮生长因子(Vascularendothelialgrowthfactor,VEGF)和CD34(Clusterofdifferentiation34,CD34)在人胃癌组织中的表达及微血管密度(MicrovessiIdensity,MVD)的临床病理意义。方法检测45例胃癌手术切除标本中胃癌组织和20例浅表性胃炎中VEGF蛋白的表达,用抗CD34单克隆抗体显示血管内皮细胞,对CD34阳性血管进行MVD计数。结果VEGF阳性表达产物主要见于细胞的胞浆内。胃癌组织中VEGF阳性表达率高于浅表性胃炎(P<0.05)。胃癌组织和浅表性胃炎中血管内皮细胞胞浆呈棕黄色染色。胃癌组织MVD值高于浅表性胃炎(P<0.05)。MVD值与TNM分期、淋巴结转移、浸润深度有关(P<0.05),与分化程度无关(P>0.05)。VEGF表达阳性MVD值高于表达阴性MVD值(P<0.05)。结论VEGF、CD34在胃癌组织中存在高水平表达,前者可能参与胃癌的血管形成和淋巴结转移。

The Clinical and pathological significance of expression of VEGF and CD34 in gastric carcinoma and MVD

Objective The current study was designed to investigate the expression of VEGF and CD34 in gastric carcinoma and clinical and pathological significance of MVD.Methods The expression of VEGF in surgical samples of 45 patients with gastric carcinoma and 20 patients wth superficial gastritis were determined by SP immunohistochemical staining method. Intratumoral VEGF and MVD were stained with anti-VEGF and vascular endothelial cells were displayed by anti-CD34 monoclonal antibody and positive vessels of them were made a MVD count. Results The positive expression of VEGF was stained mainly in the cytoplasm of cells. VEGF-positive expression percentage and the mean MVD in gastric carcinoma tissue was significantly higher than that superficial gastritis. There is significant correlation between the expression of the MVD and TNM stage,the invasive depth,lymph node metastasis( P <0.05).No significant correlation was found between the expression of the MVD and degree of differentiation( P >0.05).The MVD in VEGF-positive patients was significantly higher than that in VEGF-negative patients( P <0.05).Conclusion There are high expression of VEGF and CD34 in gastric carcinoma,the former may promoted the progression of lymph node metastasis and angiogenesis of gasric carcinoma.
