百会透曲鬓对脑出血急性期大鼠血清中VEGF、ICAM-1含量的影响

摘    要:目的 通过观察百会透刺曲鬓针刺法对脑出血急性期模型大鼠血清中血管内皮生长因子(VEGF)、细胞间黏附分子-1(ICAM-1)的动态变化的影响,探讨头穴透刺治疗脑出血可能的作用机制。方法 选取60只健康雄性Wistar大鼠,将其中54只随机分为模型组、模型+针刺组(针刺组)、模型+茴拉西坦组(西药组)3组。每组再分为出血后1 d、3 d和7 d3个时间点,每个时间点随机分配6只大鼠,制备脑出血(ICH)大鼠模型,剩余6只大鼠作为备用。采取酶联免疫吸附法(ELISA)测定血清中VEGF和ICAM-1水平。结果 针刺组与模型组相比,大鼠血清内VEGF在3 d、7 d均有显著上调(均P<0.05)。针刺组与西药组相比,出血后1 d,西药组对大鼠血清内VEGF上调显著大于针刺组(P<0.05);出血后7 d,针刺组对大鼠血清内VEGF上调显著大于西药组(P<0.05)。针刺组与模型组相比,大鼠血清内ICAM-1在不同时间点均有显著下调(P<0.05)。针刺组在出血后3 d、7 dICAM-1较西药组有显著下调(P<0.05)。结论 百会透曲鬓针刺法可以调整脑出血大鼠血清中VEGF、ICAM-1的含量,是针刺百会透刺曲鬓治疗脑出血的可能机理之一。

Abstract:Objective To explore a possible mechanism lbr scalp point-to-point acupuncture treatment of cerebral hemorrhage by dynamically observing the effect of Baihui-to-Qubin acupuncture on rat serum contents of vascular endothelial growth thctor (VEGF) and intercellular adhesion molecule-I (ICAM-I) in the acute stage of cerebral hemorrhage. Methods Sixty healthy male Wistar rats were selected. Fifty-four rats of them randomly allocated to three groups: model, model + acupuncture (acupuncture group) and model+ Aniracetam (Western drug group). Every 6 rats of each group were randomly assigned to three time points: at 1,3 and 7 days after hemorrhage, respectively. A rat model of intracerebral hematoma (ICH) was made. The remaining 6 rats were used as standbys. Serum VEGF and ICAM-1 levels were measured using an enzyme linked immunosorbent assay (ELISA). Results Rat serum VEGF was significantly up-regulated at 3 and 7 days after hemorrhage in the acupuncture group compared with the model group (both P〈0.05). As the acupuncture group was compared with the Western drug group, rat serum VEGF was significantly up-regulated to a greater extent in the Western drug group than in the acupuncture group at I day after hemorrhage (P〈0.05) and in the acupuncture group than in the Western drug group at 7 days after hemorrhage (P〈0.05). Rat serum ICAM-1 was significantly down-regulated at different time points in the acupuncture group compared with the model group (P〈0.05). ICAM-1 was significantly down-regulated at 3 and 7 days after hemorrhage in the acupuncture group compared with the Western drug group (P〈 0.05). Conclusions Baihui-to-Qubin acupuncture can regulate the serum contents of VEGF and ICAM-I in rats with cerebral hemorrhage. It is one of the possible mechanisms by which Baihui-to-Qubin acupuncture treats cerebral hemorrhage.
