SSBP1在肝癌组织中的表达及意义

目的 观察线粒体单链DNA结合蛋白(SSBP1)在肝癌组织的表达及意义.方法 收集2013年12月至2016年12月手术切除的肝癌患者组织标本80例,采用Real time-PCR和免疫组化SP法检测80例肝癌组织和癌旁组织SSBP1 mRNA和蛋白表达状况,分析其与临床病理参数的关系,并检测SSBP1在肝癌细胞株HepG2、MHCC97H、SMMC7721、Huh7及正常人肝细胞L-02的表达状况.结果 SSBP1在肝癌组织和癌旁组织的表达水平和阳性表达率分别为1.87±0.23、0.36±0.05和80.00%(64/80)、15.00%(12/80),SSBP1在肝癌组织的表达水平和阳性表达率显著高于癌旁组织,差异有统计学意义(P<0.05).HepG2细胞中SSBP1表达水平显著高于MHCC97H、SMMC7721、Huh7及L-02细胞,差异有统计学意义(P<0.05).且SSBP1表达与肿瘤大小、AFP、TNM分期、分化程度及有无门静脉癌栓、淋巴结转移密切相关(P<0.05).结论 肝癌组织和细胞系高表达SSBP1,与临床病理参数有关,且SSBP1高表达提示预后不良.

Expression and significance of SSBP1 in hepatocellular carcinoma tissues

Objective To investigate the expression and significance of mitochondria single-strand DNA protein 1 (SSBP1) in hepatocellular carcinoma tissues.Methods The primary hepatocellular carcinoma tissue samples from 80 patients who were admitted and treated by surgical ablation in our hospital from December 2013 to December 2016 were collected.The expression levels of SSBP1 mRNA and protein in 80 cases of hepatocellular carcinoma tissues and cancer adjacent tissues were detected by Real time-PCR and immunohistochemistry (SP).The correlation between SSBP1 expression levels and clinicopathological parameters of hepatocellular carcinoma was analyzed.moreover the expression levels of SSBP1 in hepatocellular carcinoma cell strains including HepG2, MHCC97H, SMMC7721,Huh7 as well as normal hepatocellular line-L-027 were detected.Results The expression levels and positive expression rate of SSBP1 in cancer tissues and adjacent tissues were 1.87±0.23,0.36±0.05 and 80.00% (64/80),15.00% (12/80), respectively,and the differences were statistically significant (P<0.05).The expression levels and positive expression rate of SSBP1 in cancer tissues were obviously higher than those in cancer adjacent tissues (P<0.05).Moreover the expression levels and positive expression rate of SSBP1 in HepG2 cells were significantly higher than those in MHCC97H,SMMC7721,Huh7 and L-027 cells (P<0.05).Furthermore the expression levels of SSBP1 were closely correlated to tumor size, AFP,TNM clinical stage,differentiation degree,portal vein tumor thrombus and lymph node metastasis (P<0.05).Conclusion The overexpression of SSBP1 exists in hepatocellular carcinoma tissues and cell strains,which is related with clinicopathological parameters of hepatocellular carcinoma,moreover, the high-expression of SSBP1 is closely correlated to poor prognosis of patients.
