Skp2蛋白在喉鳞状细胞癌中的表达及其与p27蛋白表达的关系

目的研究喉鳞状细胞癌中Skp2和p27的表达水平以及临床病理联系,探讨其在喉鳞状细胞癌发生、发展、预后中的作用。方法采用免疫组化SP法测定58例喉鳞状细胞癌(LSCC)、12例喉黏膜不典型增生(LPL)、26例癌旁正常喉组织(NLT)中Skp2和p27的表达水平。结果Skp2蛋白在LSCC和LPL中的表达均高于正常喉组织,差异有显著性(p<0.05)。p27蛋白在LSCC中低表达(43.1%)。Skp2蛋白表达与淋巴结转移、TNM分期呈正相关,与分化程度呈负相关。Skp2与p27表达水平呈负相关。Skp2表达阳性患者的5年生存率明显降低。结论Skp2通过对p27的泛素化降解,参与LSCC的发生发展过程,Skp2和p27可作为LSCC患者判断预后的有效参考指标。

Expression of Skp2 and its Correlation with p27 in Laryngeal Squamous Cell Carcinomas

Objective To detect protein expression of skp2 and p27 in laryngeal squamous cell carcinomas(LSCC).Correlation between tumor expression of Skp2 and p27 and clinicopathological characteristics of patients with LSCC was studied.Methods the expression of Skp2 and p27 in them were examined by immunohistochemically using highly specific antibodies on formalin-fixed,paraffin-embedded tissue section from 58 LSCC,12 of laryngeal precancerous lesion(LPL) and 26 normal laryngeal tissues(NLT).Results We observed the more significantly increased expression of Skp2 in LSCC and LPL groups than those in NLT group.Overexpression of Skp2 in LSCC strongly correlated with the TNM stage,lymph node metastasis,loss of tumor differentiation and loss of p27.A significant decrease in overall survival was observed in patients expressed high Skp2 levels.Conclusion These results suggest that Skp2 are involved in p27 down-regulation and may have an important role in the development of aggressive behavior in LSCC.Overexpressing of Skp2 may be used as prognostic markers for overall survival in LSCC.
