去势大鼠血清ALP动态及骨髓细胞骨形成相关基因表达研究

摘    要:目的 了解雌性大鼠切卵去势后血清碱性磷酸酶(Alkaline phosphatase,ALP)变化规律及骨髓细胞的骨形成相关基因ALP、骨钙素(Bone Gla-containing protein,BGP)及核结合因子al(core bindingfactor 1,Cbf al)的mRNA表达。方法 6月龄SD雌性大鼠18只,随机分为假切卵(Sham)和切卵(OVX)组。其中每组5只大鼠于术后起每隔10d断尾采血一次至90d,用对-硝基苯磷酸盐(P-NPP)法测量血清ALP;每组4只大鼠于术后10d处死获取骨标本,用RT-PCR方法检测骨髓细胞中ALP、BGP、Cbfal mRNA表达改变。结果 OVX组大鼠血清ALP水平,10d时已较Sham组显著升高(P<0.05, 40.0%),20d时升高呈峰值(P<0.05, 63.6%),30d时则显著性降低(P<0.05,-26.1%),40d起呈回升,与Sham组比较40、50、60、70、80及90d时的ALP分别增加26.2%(P<0.05)、18.1%、16.9%、68.5%(P<0.05)、42.6%(P<0.05)与45.9(P<0.05);切卵后10d骨髓细胞中ALP、BGP、Cbfa lmRNA的表达OVX组明显高于Sham对照组(P<0.05)。结论 大鼠去势后早期血清ALP呈先增高后下降的波动性变化,以后回升呈不同程度增高;骨髓细胞的ALP,BGP和Cbf almRNA表达呈增高改变。

Abstract:Objective To study the changes of osteogeneris-related genes in marrow cells of ovariec-tomized rats in serum alkaline phosphatase and mRNA expressions. Methods Eighteen 6-month old rats are di-vided randomly into 2 groups: ovariectomized group and sham operated control group. Five rats blood was obtained via their tails in each group every 10 days after operation till 90th day, and serum ALP was defermined by the PNPP method. Four rats in each group were killed and their bont samples were obtained 10 days after operation. Then the mRNA expressions: of ALP, BGP and CBFA1 in bone marrow cells were defected by RT-PCR.Results The serum ALP levels in ovariectomized group at 10 days were significantly higher than those in the sham group ( P <0.05, 40.0%) and reached the peak at 20 days (P < 0.05, 63.6%), Then significantly lowered at 30 days (P<0.05,-26.1%), and rose to present level at 40 days. At 40,50,60,70,80 and 90 days, the ALP lev-els in OVX group were higher than those in sham group by 26.2% (P < 0.05) , 18.1% , 16.9% ,22.5% ( P < 0.05),42.6%(P< 0.05) and45.9% (P< 0.05),respectively. Ten days after operation, the mRNA expression of relevant genes in ovariectiomized group were significantly higher than those in the sham group( P < 0.05) . Con-clusions Rise-fall fluctuations in serum ALP levels are observed in OVX rats in the early stage after ovariectony. The expressions of ALP, BGP and Cbfal in OVX group are hipher than those in the control group.
