摘    要:目的 探讨利用猪急性心肌梗死模型论证自体骨髓单个核细胞 (BM MNCs)移植对缺血性心脏病的治疗潜能和安全性。方法 结扎冠脉钝缘支造成急性心梗 ,分为两组 ,分别注射BM MNCs(BM MNCs组 )和生理盐水 (对照组 ) ,测量左室运动能力。 4周后观察移植区域的血管再生情况 ,并进行安全方面的评价。结果 在BM MNCs移植后 4周 ,左室等容收缩期压力最大变化速率 (LV dp/dtmax)和左室等容舒张期压力最大变化速率 (LV-dp/dtmax)与心梗 6 0min时相比明显改善 (P <0 .0 5 )。 4周后心梗边缘组织的小血管数目 ,BM MNCs组明显多于对照组 (P <0 .0 5 )。BM MNCs治疗后 4周未发现心电图和心肌酶的异常变化 ,心肌组织学检查也未发现炎症细胞浸润、成骨或钙化等异常现象。结论 BM MNCs移植可明显促进缺血心肌的血管再生 ,改善心肌舒缩功能

Abstract:Objective This preclinical study was designed using a swine model to investigate the safety and therapeutic potency of local bone marrow mononuclear cells (BM MNCs) implantation.Methods The swin acute coronary occlusion model was created by ligating the obtuse marginal (OM).Either BM MNCs(n=6,BM MNCs group) or normal saline (n=6,Control group) were injected into infarction and marginal areas.Hemodynamics were evaluated before and 4 weeks after the treatment.Neovascularization was evaluated by vessel count 4 weeks after the treatment.The safty of BM MNCs treatment was also evaluated.Results 4 weeks after implantantation,LV-dp/dt max and LV dp/dt max were improved in BM MNCs group ( P <0.05).Microveselus were significantly higher in the marginal area in the BM MNCs group than that in the control group 4 weeks after the treatment ( P <0.05).No significant differences in the level of TnI and no signigicant changes in ECG have been observed.No other significant histologic changes were observed in the hematoxilin eosin (HE) stained specimens from the myocardiam 4 weeks after BM MNCs treatment.Conclusions After BM MNCs treatment,the number of microvessels was increased in the ischemic myocardium,and heart function was improved.
