异丙酚预处理对大鼠脑缺血损伤不同脑区单胺类神经递质的影响

摘    要:目的观察异丙酚预处理对大鼠大脑皮层、海马及纹状体区单胺类神经递质中NE,DA的影响。方法健康Wistar大鼠24只,共分3粗:假手术组、缺血模型组和异丙酚预处理组。采用结扎双侧颈总动脉合并血压下降法造成大鼠短暂性脑缺血损伤模型,缺血30min后大鼠断头处死分离各脑区,荧光分光光度计测定各组不同脑区NE,DA的含量。结果与对照组相比,缺血模型组在大脑皮层区DA的含量增加而NE的含量没有改变;在海马区NE,DA的含量均增加;在纹状体区DA的含量增加而NE的含量没有改变,异丙酚预处理组可降低皮层区DA含量的增加及海马区NE含量的增加。结论异丙酚预处理对短暂性大鼠脑缺血损伤保护作用显著,对脑内不同脑区NE,DA的改变有一定的脑区特异性及受体选择性。

Abstract:Objective To observe the changes of NE and DA in cerebral cortex,hippocampus and striatum by preconditioning of propofol during the transient cerebral ischemia injury in rats. Methods The model of transient cerebral ischemia injury was made by the liga- tion of bilateral carotid arteries combining the descending of the blood pressure. Twenty-four male Wistar rats were stochastically divided into the sham operated group ( SH ), the ischemia group ( ISC ) and the propofol preconditioning group ( PPC ) ( n = 8 for each group). The indexes of above 3 groups were tested after 30 minutes in cerebral ischemia and anoxemia. The changes of DA and NE in different encephalic regions: cerebral cortex,hippocampus,and striatum tested by spectrophotometer(SPF). Results SPF test showed that there was a significant difference of the contents of NE and DA in cerebral cortex,hippocampus and striatum. In cerebral cortex,the content of DA in ISC group was increased ( P 〈 0.01 ) , but the content of NE was no change. PPC group could decrease the content of DA ( P 〈 0.01 ). In hippocampus, the contents of NE and DA were increased in ISC group(P 〈 0.01 ). Compared with ISC,PPC decreased NE(P 〈 0.01 ) but had no effect to DA. In striatum,the content of DA increased( P 〈 0.01 ) not the same to NE,PPC group had no effect to them. Conclusion The preconditioning of propofol had significant protection in rats with transient cerebral ischemia injury. The alterations of NE and DA in different regions by the preconditioning of propofol had the specificity of encephalic regions and the selectivity of receptors in some degrees.
