HPLC法测定清火片中大黄素和大黄酚的含量及含量均匀度

目的：建立用HPLC法测定清火片中大黄素和大黄酚的含量及含量均匀度的方法。方法：采用phenomenex HyperClone柱（C18，4．6×250mm，5μm），以流动相为甲醇-0．1％磷酸溶液（85：15），流速为1．0ml·min-1，检测波长为254nm，进样量为20μl；柱温为室温。结果：大黄素和大黄酚分别在0．5～5．5μg·ml-1（r=0．9995）和5～50μg·ml-1（r=0．9992）范围内，进样浓度与峰面积呈良好线性关系，大黄素和大黄酚的平均回收率分别为96．8％和99．4％，RSD为0．36％和0．48％（n=5）。结论：该方法简便、准确，可作为清火片质量控制的方法。

Determination of Content and Content Uniformity of Emodin and Chrysophanol in Qinghuo Tablets by HPLC

Objective： To establish method for determining the content and uniformity of Emodin and Chrysophanol in Qinghuo Tablets by HPLC. Methods： The Phenomenex HyperClone （C18, 4. 6 × 250mm, 5μm） was used and mobile phase was methanol -0. 1% Phosphoric acid （85： 15） , and its temperature was used at room temperature, with the flow rate of 1. 0ml·min-1and the wavelength of detection of 254 nm, and the injection amount of 201xl. Results： The linear ranges of emodin and chrysophanol were at 0. 5 - 5.5μg·ml-1 （ r = 0. 9995 ） and 5 - 50μg·ml-1（ r = 0. 9992 ）, the average recoveries （ n = 5 ） were 96. 8% and 99.4% with RSD 0. 36% , 0. 48%. Conclusions： The method was simple, accurate, and can be used for the quality control of Qinghuo tablets.
