摘    要:目的制备重组人骨形成蛋白-2(rhBMP-2)聚氰基丙烯酸正丁酯纳米微球缓释系统,检测其对兔骨髓间充质干细胞(BMSCs)的生物学效应。方法采用改良的乳化溶液聚合法制备rhBMP-2纳米微球;采用MTT法检测微球对BMSCs增殖的影响;碱性磷酸酶(ALP)检测试剂盒检测细胞ALP活性。结果该微球缓释系统具有一定的生物活性,可促进BMSCs的增殖,并呈剂量依赖性。其效应高于单纯施加rhBMP-2的效应。结论纳米材料是一种较好的缓释系统,作用效果优于单纯使用rhBMP-2。

Abstract:Objective To Prepare slow-release system of recombined human bone morphogentic protein-2(rhBMP-2) combined with and detect its the biological effects on rabbit bone marrow mesenchymal stem cells(BMSCs).Methods poly-emulsionized solution was used to prepare the nanometer particle of rhBMP-2,the effects of Nanometer Particle on the proliferation of BMSCs were detected by MTT and the activities of alkaline phosphatase(ALP) were detected by ALP test kit.Results The slow-release system established possessed the biological activity to enhance the proliferation of BMSCs and its effects is dose dependent.Conclusion The effects of slow-release system of rhBMP-2 combined with nanometer particle is better than that of rhBMP-2 alone.
