阿托伐他汀对急性缺血心肌细胞的保护作用

目的探讨不同剂量的阿托伐他汀预处理对大鼠急性缺血心肌细胞保护作用及其机制。方法雄性SD大鼠40只,随机分为阿托伐他汀高剂量组(20 mg.kg-1.d-1),中剂量组(10 mg.kg-1.d-1),低剂量组(2 mg.kg-1.d-1),对照组(等量生理盐水),每组10只。伊文蓝和TTC染色法确定缺血和梗死心肌的范围;比色法测定心肌组织内的髓过氧化物酶(MPO)、心肌内NO活性;酶动力法测定肌酸激酶同工酶(CK-MB)的活性。结果与对照组比较,阿托伐他汀高、中剂量组可明显减少心肌梗死面积,明显降低CK-MB和心肌内的MPO水平(P<0.01);显著增加心肌内的NO水平(P<0.05)。阿托伐他汀高、中、低剂量组间无明显差异(P>0.05)。结论高、中剂量的阿托伐他汀对急性缺血心肌细胞具有保护作用,机制可能与NO有关。

Protective effects of pretreatment with atorvastatin on acute ischemic cardiomyocytes in rats

Objective To investigate the beneficial effects and mechanism of short-term pretreatment with different dosages of atorvastatin on acute ischemic cardiomyocytes in rats.Methods Forty SD rats were randomly divided into four groups with 10 rats each:control group,low dosage of atorvastatin pretreament group(2 mg·kg-1·d-1),middle dosage of atorvastatin pretreament group(10 mg·kg-1·d-1),and large dosage of atorvastatin pretreament group(20 mg·kg-1·d-1).The infarct size and area of ischemia were determined by Evans blue and TTC staining.The levels of myocardial nitrogen monoxide(NO) and myeloperoxidase(MPO) were measured by colorimetric method.The activity of CK-MB was measured by antienzyme dynamic assay.Results Compared with control guoup,the infart size,the level of myocardial MPO and the level of serum CK-MB were decreased significantly in the large and middle dosage of atorvastatin groups(P<0.01).The level of myocardial NO was increased in the large and middle dosage of atorvastatin groups(P<0.05).There was no significant difference between the low,middle and large dosage groups.Conclusion Pretreatment with atorvastatin(10-20 mg·kg-1·day-1) has protective effect on the acute ischemic cardiomyocytes.NO is likely involved in the protective mechanism.
