Abstract:Mouse embryo implantation involves the invasion of the secondary trophoblast giantcells (STGCs) of the ectoplacental cone (EPCi into the uterine decidua , which deposits an extracellularinatrix (ECM) with abundant amounts of laminin (LN) in response to.implanting embryo. We inter-fered trophoblastic cell surface β1 , 4-galactosyltransferase (GalTasel binding to its N-acetylglucosamine(GlcNAc) residues associated with laminin oligosaccharides with a-lactalbumin, antibovine milk Gal-Tase, GlcNAc and pregalactosylation of LN, and found that these perturbations of the enzyme :sub-strate complex had no effect on EPC attachment , while prevented EPC outgrowth and STGCs migrationfrom EPC. The results indicated that GalTase mediates EPC outgrowth and STGCs migration, al-though doesn’t involve in the initial attachment of EPC on LN.
