苦参碱对人肝癌细胞及正常肝细胞增殖和粘附功能影响的比较

摘    要:目的观察苦参碱对人肝癌细胞BEL-7404及正常肝细胞QSG-7701增殖和粘附能力的影响。方法采用MTT法测定苦参碱对BEL-7404和QSG-7701的增殖;倒置相差显微镜观察细胞形态学变化;粘附实验检测细胞粘附率。结果苦参碱1.0~4.0g/L能有效抑制BEL-7404细胞的增殖。0.5~1.0g/L浓度的苦参碱能有效地诱导BEL-7404分化。当苦参碱的浓度低于0.5g/L时,其对QSG-7701细胞的增殖有一定的促进作用。0.25、0.5、0.75、1.0g/L苦参碱处理72h后,BEL-7404细胞黏附抑制率分别为18.6%、24.4%、30.3%、44.7%,与对照组比较差异均有显著性(P〈0.01);而QSG-7701细胞黏附抑制率分别为1.7%、3.5%、6.1%、37.8%,只有1.0g/L浓度的苦参碱与对照组比较有差异显著性(P〈0.01)。结论苦参碱能有效抑制肝癌细胞BEL-7404的增殖,且抑制作用具有时间、剂量依赖性;低浓度能有效地诱导肝癌细胞分化;高浓度的苦参碱有较强的抑制人正常肝细胞增殖作用,低浓度具有一定的生长刺激作用;苦参碱抑制BEL-7404细胞粘附。

Effect of Matrine on cultured human hepatocytes and hepatoma carcinoma cells in proliferation and adhesion

Abstract:Objective To investigate the inhibition effect of Matrine on the proliferation and adhersion of hepatoma carcinoma cells BEL-7404 and cultured human hepatocytes QSG-7701.Methods MTT assay was used to determine the proliferation inhibition by Matrine to BEL-7404 cell and QSG-7701 cell; to observe the rnorphocytology by inverted phase contrast microscope;Adhesive ratio was detected by adhesion assay. Results Matrine (1.0- 2.0g/L )can inhibit the proliferation of BEL-7404 cells. Matrine (0.5- 1.0g/L ) can induce differentiation of BEL-7404 cells.When the matrine concentrations was less than 0.5g/L,it could promote the proliferation of QSG-7701 cells.After BEL-7404 cells were treated with Matrine of 0.25,0.5,0.75,1.0g/L for 72h respectively,adhesive ratios were18.6%,24.4%,30.3%,44.7% respectively, Compared with that of the control group, the adhesive ratio of all groups was significantly different(P〈0.01); After QSG-7701 cells were treated with Matrine of 0.25,0.5,0.75,1.0g/L for 72h respectively, adhesive ratios were 1.7% ,3.5% ,6.1%,37.8% respectively,Compared with that of the control group, the adhesive ratio of 1.0 g/L group was signifi- cantly different (P〈0.01). Conclusion Matrine has a significant inhibiting effect on human hepatocareinoma cell proliferation in a dose-and time-dependent manner;Low concentrations can effectively induce hepatoma cell differentiation;High concentrations of Matrine can strongly prohibit human liver cell proliferation, and low concentration has a certain growth-stimulating effect; Matrine inhibit BEL-7404 cell adhesion.
