油酸钠与紫外线联用灭活血浆中病毒的研究

摘    要:采用细胞感染法,对油酸钠与短波紫外线(UVC)联用灭活血浆中病毒进行了研究。结果,3200μW/cm2 UVC与3.0mg/ml油酸钠联合作用30min或3200μW/cm2 UVC与2.0mg/ml油酸钠联合作用40min,可使血浆中水泡性口炎病毒(VSV)滴度下降 6.33~6.44lg TCID50,辛德比斯病毒滴度下降6.09—6.34lg TCID50。且两者联用协同系数(T/E)远大于1,有明显增效作用。但该方法使M13噬菌体和f噬菌体滴度下降≤3lg PFU。用 RT-PCR扩增 VSV编码 N蛋白的一段核酸,研究此消毒方法对VSV核酸的影响。结果,经UVC照射的含VSV血浆目的片段扩增阴性,油酸钠处理的含VSV血浆扩增阳性。表明UVC照射作用于VSV核酸,使之断裂,从而灭活病毒,而油酸钠对VSV的这段核酸无破坏作用。

Abstract:Inactivation of virus in plasma by combination of sodium oleate and short-wave ultraviolet rays(UVC) was studied using cell infection method. The results showed that expo- sure to combination of 3200 μW/ cm2 UVC and 3.0 mg/ ml sodium oleate for 30 min or 3200μW/ cm2 UVC and 2.0 mg / ml sodium oleate for 40 min reduced the titer of vesicular stomatitis virus(VSV) in plasma by 6.33-6.44 lg TCID50 and that of Sindbis virus(SV) by 6.09-6.34 lg TCID50. The synergetic coefficient(T/E) of these two inactivating agents was higher than I with obvious synergetic effect. However, this method reduced the titers of M13 bacteriophage and f2 bacteriophage by≤31g PFU. A segment of the nucleic acid encoding N protein of VSV was amplified with RT-PCR to study the effect of this disinfecting method on VSV nucleic acid. The result showed that amplification of the target segment in the plasma containing VSV exposed to UVC was negative, while amplification of the plasma containing VSV treated with sodium oleate was positive, indicating that UVC irradiation caused break- age of VSV nucleic acid and thus inactivated the virus, while sodium oleate had no destructive effect on this segment of nucleic acid of VSV.
