釉基质蛋白对人牙周膜成纤维细胞矿化相关蛋白的影响

摘    要:目的通过检测釉基质蛋白(Enamel matrix proteins,EMPs)作用于体外培养牙周膜成纤维细胞,对细胞骨涎蛋白、骨桥蛋白、骨粘连素、牙骨质附着蛋白(Cementum attachment protein,CAP)表达及碱性磷酸酶(alkaline phosphatase,ALP)活性的影响,探讨EMPs促进牙周组织再生的作用机制。方法酶消化法获得人牙周膜成纤维细胞(Human periodontal ligament fibroblast,hPDLF)用100mg/L的EMPs诱导,镜下逐日观察细胞形态变化,在3天、7天两个时间点利用免疫组化方法和图像分析技术检测和分析诱导条件作用后细胞骨涎蛋白、骨桥蛋白、骨粘连素表达的影响,检测细胞牙骨质附着蛋白的表达。利用ALP法检测诱导后细胞成骨活性的变化。结果 100mg/L的EMPs对hPDLF具有上调骨涎蛋白、骨桥蛋白、骨粘连素的作用,随时间延长这种促进作用愈明显;EMPs作用hPDLF 3天即表达牙骨质附着蛋白;EMPs可促进hPDLF碱性磷酸酶活性的增强。结论 EMPs在一定浓度下可促使hPDLF向成骨、成牙骨质样细胞分化。
