人膀胱癌耐药细胞系T24/ADM的建立及其耐药机制

摘    要:目的建立人膀胱癌耐药细胞系,对其耐药机制进行初步探讨。方法采用阿霉素浓度梯度递增诱导法,建立多药耐药细胞系T24/ADM。相差显微镜观察细胞,用MTT法检测该耐药细胞株对多种化疗药物的多药耐药性,应用RT-PCR和Western blot比较两种细胞P-GP、MRP和LRP的表达差异来探讨可能的耐药机制。结果建立了T24/ADM耐药细胞系,对ADM、VCR和VP16有交叉耐药,P-GP、MRP和LRP在耐药细胞系中表达升高。结论 T24/ADM具有明确的多药耐药性,其多药耐药相关基因表达升高是T24/ADM获得耐药的主要机制之一。
