不同浓度胰岛素作用的Ishikawa细胞中白血病抑制因子(LIF)的表达研究

摘    要:目的探寻高胰岛素(INS)对子宫内膜腺上皮细胞白血病抑制因子(LIF)表达的影响,探讨多囊卵巢综合征(PCOS)常见的胰岛素抵抗(IR)对其内膜容受性的影响。方法体外培养高分化子宫内膜腺癌细胞Ishikawa,分别采用浓度为10mU/L、30mU/L、90mU/L的INS溶液作用细胞24h,使用酶联免疫吸附法(ELISA)检测各浓度组细胞培养上清液中LIF水平。结果对照组LIF表达水平均高于各INS作用组;不同浓度INS作用组间无差异。结论不同浓度INS溶液作用的Ishikawa细胞培养上清液中LIF均呈低水平表达,提示IR可能成为影响PCOS患者内膜容受性因素之一,造成PCOS患者不良的生殖结局。

Abstract:Objective To study the effect of hyperinsulinemia(HI) on expression of leukemia inhibition factors(LIF) in endometrial epithelial cells and the effect of insulin resistance(IR) in polycystic ovarian syndrome(PCOS) on the endometrial receptivity. Methods Well- differentiated human endometrial adenocarcinoma cells(Ishikawa cells) were cultured in vitro. Insulin was added to the Ishikawa cells cultured with insulin at different concentrations(10mU/L, 30mU/L, and 90mU/L) for 24h. LIF levels in supernatant of cultured Ishikawa cells were measured by ELISA. Results The expression level of LIF was higher in control group than in different insulin treatment groups(F--23.70, P〈0.01). However, no significance was observed in the expression level of LIF in Ishikawa cells between different groups after treatment with insulin at different concentrations(P〉0.05). Conclusion The expression level of LIF in supernatant of Ishikawa cells cultured with insulin at different concentrations is low, indicating that the IR may be one of the factors affecting the endometrial receptivity of PCOS, thus leading to the worse outcome in PCOS patients.
