动物疟原虫与人疟原虫的交叉反应抗原

摘    要:目的分析5种疟原虫(间日疟原虫、恶性疟原虫,食蟹猴疟原虫、伯氏症原虫,约氏疟原虫)中,可被间日疟、恶性疟病人血清识别的交及反应抗原。方法用50%、60%Percoll不连续梯度分离疟原虫感染红细胞,20mmolPBS被红细胞后,收集红内期原虫,以1%NP—40提取可溶性抗原。各种抗原经SDS—PAGE电泳后,转移至硝酸纤维膜上,分别用间日疟、恶性疾病人血清进行免疫识别。结果3神动物疟原虫红内期抗原中有多条蛋白区带可被间日疟或恶性疟病人血清识别。其中合蟹报疟原虫有14条蛋白带被间日疟病人血清识别,伯氏疟原虫有16条蛋白区带被恶性疟病人血清识别,分别多于其它疟原虫。间日疟病人血清可识别5种疟原虫共有的72kD蛋白,而恶性疟病人血清不识别该蛋白。结论人疟原虫和动物疟原虫之间有一系列支又反应抗原。会压滤疟原虫与间日疟原虫,伯氏疟原虫与恶性疟原虫之间抗原相似程度高于其它疟原虫。72kD.蛋白在间日疟无疾学诊断方面可能有实用价值。

Abstract:To analyze the blood- stage anti -gens from Plasmodium vivax , P.falciparum, P.cynomolgi, P. berghei and P. yoelu which could be recognized by the sera from P. vivax and P.falciparum malaria patients.Methods Erythrocytes infected with the five species of Plasmodia were isolated by 50%, 60%Percoll density gradient. After the erythrocytes were lysed by 20 mmol PBS, the released parasites were collected and soluble antigens were extracted with the buffer including 1% NP-40. Using the sera from P- vivax and P. falciparum malaria patients as probs, the above antigens were detected by means of western blot.Results A series of the protein bands from different species of Plasmodia could be recognized by the sera of P. vivax or P, falciparvm malaria patients. The 14 protein bands from P. cynomolgi could be recognized by the sera from P. vivaxmalaria patients and the 16 protein bands from P.berghei could be done by the sera from P. falciparum malaria patients. That were more than these of the other plasmodia. The protein bands with molicular weight of 72kDa,which was common among the five species of Plasmodia, coud be only recognized by the sera from P. vivax malaria patients.Conclusion There is a series of cross-reacting antigens among the five species of Plasmodia.There a higher similarity between P. vivax and P.cynomolgi or between P. berghei and P. Falciparum than the other Plasmodia. 72kDa protein is perhaps valuble in immunologic diagnosis of P. vivax malaria.
