人骨髓中自然抑制细胞的抑瘤活性

摘    要:目的研究人骨髓细胞中自然抑制细胞的抗肿瘤效应。方法分离并培养人骨髓中不同的细胞亚群,将不同的骨髓细胞亚群或其培养上清加入HL-6O或Kato-3肿瘤细胞培养体系中,检测肿瘤细胞的增殖反应,并检测电离辐射及蛋白质合成阻断剂对骨髓自然抑制活性的影响。结果具有自然抑制活性的骨髓细胞主要为CD34+CD33-细胞,其不但可以抑制丝裂素原刺激的人外周血单个核细胞的增殖,而且可以抑制肿瘤细胞株的增殖。自然抑制细胞发挥其自然抑制活性不需要细胞接触,但是蛋白质的合成则是这一过程所必需的,因为嘌呤霉素阻断自然抑制细胞发挥其自然抑制活性。此外,我们的结果表明,自然抑制细胞可分泌一种可溶性因子,IL-3和GM-GSF可以促进这一可溶性因子的产生。结论人骨髓细胞中自然抑制细胞具有显著的抗肿瘤效应。

Abstract:To investigate the inhibition of tumor cell proliferation by natural suppressor (NS) cells in human bone marrow.Methods Different subpopulations of human bone marrow cells were isolated and cultured, and then the subpopulation or its culture supernatant was added into the culture system of HL - 6O or Kato - 3 cell lines. The tumor cell proliferation was assayed-.Effect of irradiation or protein synthesis blockade on bone marrow natural suppressor activity was investigated.Results In human bone marrow, NS cells were CD34 CD33-cells. NS cells. libited not only the proliferation of human mononuclear cells in peripheral blood which had been stimulated by mitogens but also the proliferation of tumor cell lines- Cell - to - cell contact was not necessary for NS cells to exert NS activity. However, Protein synthesis was neccssary for NS eells to suppress Proliferation, since puromycin blocked NS activity. In addition, NS cells were found to secrete a factor or factors. The secretion was enhanced beIL - 3 and GM - CSF.Conclusion Naturral suppressor cells in human bone marrow occupied a significant anti - tumor activity.
