聚乳酸粒子异基因抵抗抑制作用的研究

摘    要:目的研究策乳酸籽子(PLAM)对异基因抵抗作用的抑制作用。方法于异基因骨髓移植前不同时间静脉络以不同剂量PLAM,测定移植受体的存活时间以及移植受体day9和day14CFU-S的形成.结果移植前胸静脉注射PLAM:0.75g·kg-1体全可明显促进移植受体day9和day14CFU-S的形成,同时明显延长移植受体的存活时间,而低于该剂量或在移植前2或24h静脉注射PLAM则无效,当静脉注射PLAM1g·kg-1体重以上时,则可同时促进内源性CFU-S的形成。结论吞噬细胞与异基因骨髓移植时存在的异基因抵抗效应密切相关;一定剂量的PLAM于移植前一定时间静脉注射给移植受体可明显减轻异基因抵抗效应,延长移植受体的存活时间;大剂量PLAM静脉注射可提高机体的辐射抵抗能力。

Abstract:To investigate the inhibitory activity of poly - (1-lactic acid) microspheres (PLAM) on the allogeneic resistance Methods Various doses PLAM were admininstrated via system vein at different tines before allogeneic bone marrow transplantation (alloBMT). And then,the survival time and day9 CFU -S and day14 CFU-S of the recipients were assayed.Results The results shown that venous injection of PLAM 0. 75g. kg-1 6 hours before transplantation promoted the day 9 CFU-S and day 14CFR-S formation of the transplanted recipients,and at the same time,resulted in the great improvement of the survival of the recipients. When the PLAM dose was less than 0. 75. kg-1 or PLAM injected 2 hours or 24 hours be fore alloBMT, there were no effective. When the PLAM dose was much more than 1g. kg-1, there wereendogenous CFU-S formation in the recipients.Conclusion Based on thesc results, we concluded that phagocytic cells played an essential role, probably as effector cells in genetic resistance to bone marrow allograft. The results also indicated that PLANI administration in an appropriat dose at appropriat time was able to suppress genetic resistance to bone marrow allograft and a big dose of PLAM administration increased the ability of radiation resistance of animals.
