甲基苯丙胺与蛋白质结合物的合成

摘    要:目的:利用蛋白质连接法合成甲基苯丙胺人工全抗原。方法:以BSA蛋白做载体蛋白,用碳二亚胺法偶联合成甲基苯丙胺抗原,并用紫外扫描、蛋白电泳以及胶体金免疫层析等方法进行检测。结果:甲基苯丙胺与BSA连接成功,连接产物分子量增加,且每分子牛血清白蛋白连接的甲基苯丙胺分子数为21个;胶体金免疫层析检测结果显示,合成抗原与抗体有特异性反应,具有活性,交叉反应良好。结论:此抗原可作为甲基苯丙胺免疫用抗原。

Abstract:Objective:To synthesize methamphetamine antigen by conjugating methamphetamine to protein.Methods:Methamphetamine antigen was synthesized by EDC method,and the carrier protein was BSA.The reaction was monitored by UV scanning,protein electrophoresis and GICA methods.Results:The conjunction was successful and the molecular weights of protein conjugates increased,and 21 molecules of methamphetamine were conjugated to one molecule of BSA,and the formed conjugate showed confirmed antigenic activity and well cross reaction.Conclusion:Methamphetamine antigen is successfully synthesized and could be used as immunity antigen.
