电针治疗抑郁症的全基因组表达谱芯片数据分析

摘    要:目的:研究电针治疗抑郁症的机制。方法:采用Open-Field法作行为学测试,选取活动度相近的30只大鼠进行实验,随机分为正常组、模型组、电针组,每组10只。模型组、电针组应用慢性不可预见性应激(冰水游泳、电击、夹尾等)合并孤养(持续21 d)的方法造成大鼠抑郁症模型。电针组选择"百会""印堂"穴电针15 min(频率2 Hz,输出电流1 mA),1日1次,连续7 d。分别于造模前、造模结束时和治疗1周后测定各组大鼠的行为学(水平和垂直方向的运动)和1%蔗糖溶液消耗量。取各组大鼠的海马组织进行Roche-Ni mbleGen全基因组表达谱芯片测试并进行数据分析。结果:造模后,大鼠行为学能力和1%蔗糖溶液消耗量均明显降低(P0.01);电针后,上述指标均有改善趋势,其中1%蔗糖溶液消耗量的改善具有统计学意义(P0.05)。基因芯片测试结果显示,抑郁症模型大鼠下调表达,而经电针治疗后表达趋于正常的基因有101条,其中多条基因与能量和物质代谢有关,还涉及蛋白质生物合成和嗅觉感受器等;抑郁症模型大鼠上调表达,而经电针治疗后表达趋于正常的基因有125条,其中多条基因与凝血、免疫、炎性反应和凋亡相关。结论:电针治疗抑郁症的机制与调控多条基因表达有关。
