气体信号分子硫化氢在大鼠肝缺血-再灌注损伤中的作用

摘    要:目的探讨气体信号分子硫化氢(H2S)在大鼠肝缺血-再灌注损伤(HIRI)过程中的作用机制。方法将56只健康雄性SD大鼠随机分为7组(每组8只),分别给予不同的处理。采用Pringle法制作HIRI模型。采用敏感硫电极法测定各组血清H2S的含量;原位细胞凋亡法测定肝细胞凋亡指数。全自动生化分析仪测定血清ALT和ALP含量;丙二醛、谷胱苷肽测定试剂盒测定肝组织MDA、GSH含量。结果各实验组H2S、ALT、ALP、MDA和AI水平较SO组明显升高,GSH水平则明显下降,组间差异显著(P<0.05)。受NaHS干预后的ALT、ALP、MDA和AI水平与相同时相组比较明显下降,H2S、GSH水平则明显升高,组间差异显著(P<0.05)。H2S与ALT、ALP、MDA、AI和GSH的相关系数分别为0.92、0.90、0.87、0.89和-0.77(P<0.01),说明H2S与其他指标高度相关。结论内源性H2S作为气体信号分子在HIRI过程中能抑制肝细胞凋亡,对肝细胞具有积极的保护作用。

Abstract:Objective To explore the role of hydrogen sulfide(H2S),gaseous signal molecule on the hepatic ischemia reperfusion injury(HIRI) in rats.Methods 56 male SD rats were divided into seven groups randomly(n=8 in each group),and were treated differently.The model of HIRI was established by Pringle's hepatic vascular occlusion.The contents of H2S in serum were measured by sensitive sulf-electrode;The contents of alanine aminotransferase(ALT) and alkali phosphatase(ALP) in the serum were measured by holo-automatic biochemistry analyzer;hepatic tissue were observed by hematoxylin and eosin stain;The contents of malondialdehyde(MDA) and reduced glutathione hormone(GSH) in hepatic tissue were measured by MDA and GSH kits;The changes of hepatic apoptotic index(AI) were measured by TUNEL in each group respectively.Results The contents of H2S,ALT,ALP,MDA and AI of every experimental group were rised obviously than group sham operation(SO),however,the contents of GSH were lowered obviously than group SO,interclass difference were significant(P<0.05).The contents of ALT,ALP,MDA and AI of each experimental group by NaHS interfered were lowered obviously than the same phase group,however the contents of H2S and GSH were rised obviously,interclass difference were significant(P<0.05).Correlation coefficient between H2S and above-mentioned index was 0.92,0.90,0.87,0.89 and-0.77 respectively(P<0.01),correlation among them were significant.Conclusion Endogenous H2S can inhibit hepatic apoptosis in HIRI in rats as gaseous signal molecule,showed significant protecting effect among the processs of HIRI in rats.
