鼻咽癌细胞的糖皮质激素受体含量及受新酮醛的影响

摘    要:以~H-地塞米松(~3H-dexamethasone,~3H-Dex)为放射配基,测定鼻咽癌细胞糖皮质激素受体(GCR)含量,结果当用地塞米松(Dex)非标记配基作竞争性试验时,GCR水平较高,而且低分化鼻咽癌上皮细胞(CNE-Ⅱ)GCR含量明显高于高分化细胞(CNE-I)(P<0.05);用新酮醛、新鱼腥草素或多糖替换Dex作竞争物时,CNE-Ⅰ细胞由于受新酮醛作用影响,GCR含量明显高于Dex作用时的含量(P<0.05);CNE-Ⅱ细胞GCR含量提示新酮醛、新鱼腥草素、Dex三者的作用大致相同(P>0.05)。此外,以CNE-Ⅰ和CNE-Ⅱ作竞争物时均测不到GCR含量,且为负值。

Abstract:This paper reports the determination of the level of glucocorticoid receptors (GCR) in CNE nasopharyngeal carcinoma cells by using 3H-dexamethasone as redioligand. The results showed that CNE-1 and CNE-2 can have a higher level of GCR, by using non labeling Dex ligand as competitor. The value of GCR in poorly differentiated nasopharyngeal carcinoma epithelial cell line (CNE-1) is significant higher than in well defferentiated nasopharyngeal carcinoma cell line (CNE-2) (P<0.05). There were some effects on the level of GCR in CNE-1 and CNE-2 by using New keto-aldehyde, Neo-Huttuynium or polyglucose as competitor instead of non labeling Dex ligand. When New keto-aldehyde was used the level of GCE in CNE-1 was signficantly higher than non labeling Dex (P <0.05). However, the level of GCR in CNE-2 has no significant defference with those after using New keto-aldehyde, Neo-huttuynium or polyglucose. GCR levels in CNE-i and CNE-2 were negative when polyglucose was used.
