^99Tc-MDP与“博宁”治疗骨质疏松动物模型分析

摘    要:目的 用同位素药物^99Tc-MDP与双膦酸盐类药物“博宁”治疗骨质疏松动物模型,比较实验室的骨密度等测定的变化。方法 由药物盐酸地塞米松诱导制作兔骨质疏松动物模型,分为正常对照组、骨质疏松对照组和骨质疏松^99Tc-MDP与“博宁”治疗组,总疗程时间为15周。处死取实验兔股骨、第3、4腰椎、股骨头部位进行骨细胞学、骨密度测定。结果 对实验兔予以肌肉注射DX6周后,正常对照组基本无骨质破坏的征象,而骨质疏松模型组则表现为股骨头关节表面软骨破坏,骨小梁稀疏、断裂,骨母细胞消失等明显骨破坏现象。说明本次实验的骨质疏松的动物模型成立。经^99Tc-MDP治疗组与博宁治疗组与骨质疏松组比较,腰椎骨密度分别上升25.90%和19.61%,股骨头骨密度上升26.38%和12.28%,核素骨显像差值上升17.32%和6.27%。结论 ^99Tc-MDP治疗由糖皮质激素诱导的兔骨质疏松模型的结果提示,^99Tc-MDP治疗骨质疏松的疗效优于“博宁”,为治疗人体由糖皮质激素引起的骨质疏松症提供了依据。

The study of ^99Tc-MDP and BONING in the treatment on the animal osteoporosis model

Abstract:Objective To compare the effect in the treatment on animal osteoporosis model by isotope drug ^99Tc-MDP with diphosphonate drug BONING by BMD examination etc. Methods After the animal osteoprosis model induced by hydrochloric dexamethasone was set up, the rabbits were separated into 4 groups, in 2 groups of them were treated respectively by isotope drug ^99Tc-MDP and diphosphonate drug BONING, and the other two groups are normal control group and osteoporosis control group. After being treated for 15 weeks, the rabbits were killed and the femur, L3, L4, and femur head of each rabbit were taken to check the bone cytology, bone mineral density and bone biomechanics. Results Intramuscularly injected with hydrochloric dexamethasone for 6 weeks, the rabbits in osteoporosis model group showed obvious signs of osseous destruction such as destruction in the facies articularis of femur head, rarefaction and fragmentation of trabecula, disappearance of myeloid cells, while those in control groop showed few sign of osseous destruction. It showed that the animal osteoporosis model in this experiment was successfully set up. After treated for 15 weeks, compared with osteoporosis control group, ^99Tc-MDP and BONING group increased in lumbar bone density for 25.90% and 19.61%, in femur head bone density for 26.38 % and 12.28 %, in radionuclide bone scintigraphy value for 17.32 % and 6.27%. It also showed some improvement in bone cytology. Conclusion The result of the study and BONING in the treatment on animal osteoporosis model induced by adrenal cortical hormone showed that the therapy effect in osteoporosis ^99Tc-MDP is better than that of BONING. The results gave some support in the reason of osteoporosis treatment.
