肝细胞癌组织中CYP27A1的表达及其临床意义

摘    要:目的:探讨CYP27A1在肝细胞癌(HCC)组织中的表达及其临床意义。方法:基于癌症基因组图谱(TCGA)数据库,利用R软件采用Wilcoxon秩和检验、配对资料Wilcoxon符号秩检验比较CYP27A1在肝细胞癌和正常肝组织中的表达情况。采用Kaplan-Meier分析及Cox回归分析评估CYP27A1和生存率之间的相关性并探索HCC独立危险因素。采用基因富集分析(GSEA)结合蛋白互作网络进行CYP27A1的生物学功能研究。结果:CYP27A1在肝癌组织中的表达显著低于正常肝组织(P<0.01);CYP27A1表达水平与HCC患者临床分期显著相关,临床分期越高,CYP27A1表达越低(P<0.01);CYP27A1低表达与HCC患者不良预后相关,并且是影响HCC患者预后的独立危险因素(P均<0.05);CYP27A1低表达样本富集到细胞周期、粘着力、胞吞作用、基因复制等功能及ErbB信号通路和p53信号通路。结论:CYP27A1表达与HCC临床分期及预后相关,可能成为临床诊治HCC的生物标志物。

Abstract:Objective:To investigate the expression and clinical significance of CYP27A1 in hepatocellular carcinoma(HCC).Methods:Based on The Cancer Genome Atlas(TCGA),We compared the expression of CYP27A1 in hepatocellular carcinoma and normal tissues with Wilcoxon rank sum test and Wilcoxon signed ranks test by R,Kaplan Meier and Cox regression was performed to evaluate the correlation between CYP27A1 and overall survival,and explore independent risk factors for HCC.Gene set enrichment analysis(GSEA)and PPIs network was also conducted to annotate biological function of CYP27A1.Results:The expression of CYP27A1 in HCC tissues was significantly lower than that in normal liver tissues(P<0.01).The expression level of CYP27A1 was correlated with the clinical stage of HCC patients,and the higher the clinical stage was,the lower the CYP27A1 expression was(P<0.01).Low expression of CYP27A1 is associated with poor prognosis in HCC patients,and is an independent risk factor affecting the prognosis(P<0.05).ErbB/p53 pathway,cell cycle,focal adhesion,endocytosis and DNA replication are differentially enriched in CYP27A1 low expression phenotype.Conclusion:The expression of CYP27A1 is correlated with the clinical stage and prognosis of HCC patients,and CYP27A1 may be used as a biomarker for the clinical diagnosis and treatment of HCC.
