消栓通络颗粒对动脉粥样硬化大鼠防治作用及机制研究

摘    要:目的研究消栓通络颗粒对动脉粥样硬化(AS)模型大鼠的防治作用及其可能的机制。方法采用高脂饲料喂养和腹腔注射维生素D建立AS模型,40只雄性SD大鼠随机分为4组,正常组(喂食基础饲料)、模型组(等量生理盐水)、消栓通络颗粒低高剂量组(1.8 g/kg和7.2 g/kg),12周后处死各组实验大鼠,取主动脉组织,测定超氧化物歧化酶(SOD)、丙二醛(MDA)、内皮素-1(ET-1)、血管内皮生长因子(VEGF)和一氧化氮(NO)水平。结果经消栓通络颗粒治疗后,低剂量组与高剂量组大鼠血管组织中过氧化物代谢产物MDA则明显减少,均有统计学差异(P<0.05;P<0.01),实验大鼠血管组织中SOD略有升高但无统计学差异(P>0.05);消栓通络颗粒高剂量组能够明显降低ET-1水平,升高VEGF水平,与模型对照组比较均具有统计学差异(P<0.05),消栓通络治疗组血管组织样本中NO水平变化,与模型对照组比较无明显差异。结论消栓通络颗粒对AS模型大鼠有一定防治作用,其机制可能与缓解组织氧化应激状态,减轻内皮损伤有关。

Preventive and therapeutic effects of Resolving-thrombus Freeing-collateral Granules in rats with atherosclerosis and a study on mechanism

Abstract:Objective To study the preventive and therapeutic effects of Resolving-thrombus Freeingcollateral Granules on atherosclerosis(AS)in model rats,and discuss possible mechanism.Methods AS model was established through giving high fat feed and intraperitoneal injection of VD in rats.Male SD rats(n=40)were divided randomly into 4 groups:normal group(basic feed),model group(the same amount of saline solution),lowdose Resolving-thrombus Freeing-collateral Granules group(1.8 g/kg,low-dose group)and high-dose Resolvingthrombus Freeing-collateral Granules group(7.2 g/kg,high-dose group).After 12 weeks,the rats in all groups were sacrificed for collecting aortic tissue,and the levels of superoxide dismutase(SOD),malondialdehyde(MDA),endothelin-1(ET-1),vascular endothelial growth factor(VEGF)and nitric oxide(NO)were detected.Results After treatment with Resolving-thrombus Freeing-collateral Granules,MDA level decreased significantly(P<0.05,P<0.01)and SOD level increased slightly(P>0.05)in vascular tissue in low-dose group and high-dose group.In high-dose group,ET-1 level decreased significantly and VEGF level increased compared with model group(P<0.05).In low-dose group and high-dose group,the change of NO level had no significant difference compared with model group.Conclusion Resolving-thrombus Freeing-collateral Granules have some preventive and therapeutic effect on AS in modeled rats,and the mechanism maybe related to alleviating tissue oxidative stress and relieving vascular endothelium injury.
