络风宁2号方对心肌梗死后心力衰竭大鼠心功能和心肌纤维化的影响

目的观察络风宁2号方对心肌梗死(心梗)后心力衰竭(心衰)大鼠心功能和心肌纤维化进程的影响及其可能的药理机制。方法采用左冠状动脉前降支结扎法制作心梗后慢性心衰模型,雄性SD大鼠56只随机分为模型组6只,卡托普利组6只,络风宁2号方低剂量组6只,络风宁2号方中剂量组7只,络风宁2号方高剂量组6只,假手术组5只,造模手术存活率86%,造模成功率70%,假手术组大鼠存活率83.3%。络风宁2号方低中高剂量组及卡托普利组连续给药4周,其他组灌服生理盐水4周后,超声心动图检测各组大鼠心脏结构及心功能、ELISA法检测大鼠血清N末端脑钠肽前体(NT-proBNP)水平、Masson染色法观察大鼠心肌纤维化情况并测算胶原容积分数(CVF)。结果和模型组比较,络风宁2号方各剂量组、卡托普利组大鼠一般情况有所改善,左室舒张末期内径(LVDd)、左室收缩末期内径(LVDs)降低(P<0.05),左室射血分数(LVEF)升高(P<0.05),血清NT-proBNP水平、CVF明显下降,差异具有统计学意义(P<0.05或P<0.01),且在一定范围内同剂量呈正相关;卡托普利组和络风宁2号高剂量组之间NT-proBNP水平和心肌细胞纤维化程度比较的差异均无统计学意义(P>0.05)。结论络风宁2号方有较好的改善心衰大鼠心功能和抗心肌纤维化的作用,且此作用在一定范围内同剂量呈正相关,同时高剂量络风宁2号方和卡托普利改善CHF大鼠心功能和抗心肌纤维化的作用相当。

Influences of Luofengning Granules II on heart function and myocardial fibrosis in rats with chronic heart failure after acute myocardial infarction

Objective To observe the influences of Luofengning GranulesⅡon heart function and myocardial fibrosis process in rats with chronic heart failure(CHF)after acute myocardial infarction(AMI),and discuss the possible medicinal mechanism.Methods The model of CHF after AMI was established by applying ligation of left anterior descending coronary artery.SD rats(n=56)were randomly divided into model group(n=6),captopril group(n=6),lowdose Luofengning GranulesⅡgroup(n=6),mid-dose Luofengning GranulesⅡgroup(n=7),high-dose Luofengning GranulesⅡgroup(n=6)and sham-operation group(n=5).The survival rate of modeling was 86%,success rate of modeling was 70%,and survival rate was 83.3%in sham-operation group.All Luofengning GranulesⅡgroups and captopril group were given corresponding drugs for 4 weeks and other groups were orally given normal saline for 4 weeks.The heart structure and function were detected by using echocardiogram in all groups.The level of serum NT-proBNP was detected by using ELISA.The status of myocardial fibrosis was observed by using Masson staining and collagen volume fraction(CVF)was calculated.Results Compared with model group,the general conditions were improved,LVDd and LVDs decreased(P<0.05),LVEF increased(P<0.05),and level of serum NT-proBNP and CVF decreased significantly(P<0.05 or P<0.01)in all Luofengning GranulesⅡgroups and captopril group,which were positively correlated to dose within certain limits.The differences in level of serum NT-proBNP and severity of myocardial fibrosis had no statistical significance between captopril group and high-dose Luofengning GranulesⅡgroup(P>0.05).Conclusion Luofengning GranulesⅡcan improve heart function and anti-myocardial fibrosis,and its effect is positively correlated to dose within certain limits.At the same time,high-dose Luofengning GranulesⅡand captopril have the similar effects on improving heart function and anti-myocardial fibrosis in CHF rats.
