| 摘 要： | 目的：观察补肾活血方对家兔膝骨关节炎（OA）滑膜组织基因表达谱的调控作用。方法：Hulth方法建立家兔膝骨关节炎模型,给予补肾活血方干预、常规抽提和纯化RNA,采用AffymetrixRat230 2.0Array芯片检测滑膜组织的基因表达变化。结果：芯片结果显示补肾活血方对模型组滑膜组织基因表达具有明显调控作用,补肾活血方有明显上调作用的基因中,中药组比模型组上调1.8倍以上的有10个;具有明显下调作用的基因中,中药组比模型组下调2倍以上的有4个。结论：补肾活血方可能通过调控目前在骨关节炎发生、发展中作用尚不明确的基因,起到抑制滑膜病变进展、促进软骨修复,从而达到治疗骨关节炎作用。

| Abstract: | Objective: To explore the regulating effect of Chinese medicine Bushen Huoxue Formula on the gene expression profile of synovium in rabbits with knee osteoarthritis.Methods: After the rabbit model of knee osteoarthritis was eatablished,Bushen Huoxue Formula was administered.Then RNA in synovium of diseased knee was extracted and purified.Gene expression was analysed by using Affymetrix genechip operating system.Results:The results showed that among the significantly up-regulated genes,10 genes of TCM group were increased 1.8 times more than that in model group.In the significantly down-regulated genes,4 genes of TCM group were decreased twice more than that in model group.Conclusion: Bushen Huoxue Formula may achieve the treatment effect on osteoarthritis by regulating genes whose roles are not clear currently in the occurrence and development of osteoarthritis. |
