杨梅树皮提取物及杨梅素抗肿瘤活性

摘    要:目的对杨梅树皮提取物、提取物的大孔吸附树脂各洗脱部位及杨梅素单体化合物进行体外抗肿瘤研究。方法采用MTT法测定细胞死亡率;采用倒置显微镜观察细胞形态学变化;用琼脂糖电泳观察DNA片段化。用Western印迹法分析药物对蛋白质水平的影响。结果杨梅树皮提取物及其大孔吸附树脂洗脱物、杨梅素单体化合物体外对人宫颈癌HeLa细胞、人黑色素瘤A375-S2细胞、人乳腺癌MCF-7细胞和人肝癌HepG2细胞均具有明显的细胞毒作用。杨梅素明显抑制HeLa细胞的增殖,诱导HeLa细胞调亡。Caspase-3、Caspase-9抑制剂可明显抑制50 mg.L-1杨梅素诱导的凋亡。Western印迹结果显示50 mg.L-1杨梅素作用36 h后Caspase-3前体及其底物ICAD和PARP发生降解。结论杨梅树皮提取物及其大孔吸附树脂洗脱物具有明显的细胞毒作用,杨梅素(50 mg.L-1)通过激活Caspase途径诱导HeLa细胞凋亡。

Abstract:Objective To study the antitumor activities of the extracts from the bark of Myrica rubra Sieb. Et Zucc. and myricetin. Methods Solvent extraction together with column chromatography was used to isolate chemical constituents. Cell viability was measured by MTT method. Morphological changes were observed by phase contrast microscopy and Hoechst 33258 staining. DNA fragmentation was assayed by agarose gel electrophoresis. Protein level was detected by Western blot analysis. Results Myricetin induced HeLa cell apoptosis. The apoptosis of HeLa cells was partially reversed by caspase-3 and caspase-9 inhibitors. Treatment of HeLa cells with myricetin for 36 h induced ICAD and PARP degration. Conclusions Myricetin induces HeLa cell death via activation of caspases.
