Survivin基因在喉癌中的表达及意义

目的 研究凋亡抑制蛋白Survivin基因在喉癌组织及喉癌细胞株中的表达,探讨其在喉癌发生发展中的作用。方法 采用免疫组织化学技术,研究42例喉癌标本、20例癌旁组织、20例喉角化症、10例正常喉黏膜中Survivin 蛋白的表达情况;并用RT-PCR技术检测喉癌细胞株Hep-2中Survivin mRNA的表达。结果 正常喉黏膜中无Survivin表达, 喉癌组织中Survivin 蛋白的表达 (66.6 %) 明显高于癌旁组织 (40.0 %)和喉角化症(35.0 %)(P<0.001),喉癌组织中Survivin表达与临床分期、病理分级及有无颈淋巴结转移无明显相关性(P > 0.05)。喉癌细胞株Hep - 2中有较强Survivin mRNA的表达。结论 Survivin基因在喉癌组织及喉癌细胞株中的高表达可能与喉癌的发生关系密切,具体机制有待于进一步研究。

Expression and implication of survivin gene in laryngeal carcinoma

OBJECTIVE To investigate the ex-pression of inhibitor apoptosis protien of survivingene and determine the role in the pathogenesisof laryngeal squamous cell carcinoma. METHODSThe expression of survivin gene were analysedby immunohistochemistry SP method in 42 la-ryngeal carcinoma tissues,20 laryngeal kerato-sis tissues,20 tissues adjacent to the cancer,and10 normal laryngeal tissue. RT - PCR was usedto test the survivin mRNA expression in humanlaryngeal squamous cell line Hep-2. RESULTSThe expression of survivin gene was not observedin normal laryngeal tissues. The positive rate ofexpression of survivin protein in laryngeal carcinoma(66.6 %) was significantly higher than that in theadjacent cancer tissues (40 %) and in keratosis oflarynx (35 % ) (P < 0.001). But the expression rateof survivin gene was not related to clinical stageand pathological grade and neck lymph nodemetastases(P > 0.05).The expression of survivin mRNAwas detected in Hep-2. CONCLUSION The up-regu-lation expression of survivin gene in laryngeal car-cinoma suggested that survivin gene may play arole in the pathway of carcinogensis.
