| 摘 要： | 本研究旨在比较人脐带羊膜来源的间充质干细胞(mesenchymal stem cells,MSC)与骨髓来源MSC的免疫调控能力,为临床开展异基因造血干细胞移植提供实验依据。采用胶原酶消化法分离人脐带羊膜贴壁细胞,并从细胞形态、生长特性、细胞表型、多向分化能力进行鉴定。利用淋巴细胞转化实验和混合淋巴细胞反应比较人脐带羊膜来源MSC与人骨髓来源MSC的免疫调控能力的差异。结果表明,人脐带羊膜来源和骨髓来源的MSC具有相似的生物学特征,即细胞呈成纤维样形态生长,并具有强大的体外扩增能力;流式细胞仪检测细胞表型结果显示,脐带羊膜来源MSC高表达CD73、CD90、CD105,而骨髓来源MSC表面标志CD34、CD45以及参与免疫反应的细胞表面分子HLA-DR和CD86等为阴性。其诱导多向分化能力结果表明,两组细胞均可向成脂肪、成骨和成软骨分化。混合淋巴细胞反应和淋巴细胞转化实验证明,两组MSC均可抑制异体T细胞的增殖,且随MSC数量的增加,抑制效应逐渐增强;RT-PCR检测显示两组MSC表达相同的免疫相关因子。结论:人脐带羊膜来源的MSC可以替代成人骨髓MSC,可作为满足实验和临床需要的MSC重要来源。
