| 硼替佐米单用或联合三氧化二砷对Jurkat细胞凋亡和Livin mRNA表达的影响 |

| 摘 要： | 本研究旨在探讨硼替佐米单用或联合三氧化二砷对Jurkat细胞凋亡的影响。用不同浓度硼替佐米、三氧化二砷处理细胞24小时,采用MTT比色法检测细胞增殖活性,流式细胞术检测细胞凋亡,RT-PCR法检测Livin mRNA的表达。结果表明,5-25 nmol/L硼替佐米对Jurkat细胞增殖均有一定抑制作用,随着浓度增加抑制作用逐渐增强。硼替佐米和三氧化二砷联合处理组对细胞的抑制作用比同剂量两药中任一种单独使用时都显著增强(p<0.05)。硼替佐米能促使Jurkat细胞凋亡,且随浓度增加而凋亡增多,联合处理组对细胞的凋亡作用比同剂量单药作用时显著增强(p<0.01)。硼替佐米能下调Jurkat细胞Livin mRNA的表达,且随着药物浓度增加,表达逐渐降低,联合处理组比同剂量单药组更显著下调LivinmRNA的表达(p<0.05)。结论:硼替佐米或联合三氧化二砷可以下调Jurkat细胞中Livin mRNA的表达,并可能通过此机制诱导K562细胞凋亡,两药具有协同作用。
