| 摘 要： | 目的 检测非小细胞肺癌患者基质溶解素（matrilysin，MMP-7）的表达，探讨重组基质溶解素（rMMP-7）和脂质体-MMP-7反义寡核苷酸（antisense oligonucleotde of matrilysin by liposome transfection，LIPO-MAON）对肺腺癌A549细胞和人脐静脉血管内皮细胞（human umbilical vein endothelial cell，HUVEC）增殖的影响。方法 ①免疫组化法检测NSCLC患者癌组织MMP-7表达；②MMT法和半定量RT-PCR法检测rMMP-7和LIPO-MOAN作用下A549和HUVEC的增殖及MMP-7-mRNA表达。结果 57例肺癌组织47例MMP-7表达为阳性，阳性表达率为82．5％，与正常对照组相比P〈0．01。rMMP-7能促进肺腺癌A549细胞的增殖，LIPO-MAON对AM9细胞增殖则有明显抑制作用。对HUVEC增殖，仅高浓度的rMMP-7（0．75、1．0μg／ml）有促进作用，而LIPO—MAON对其增殖无抑制作用。RT-PCR结果表明，HUVEC不表达MMP-7-mRNA；rMMP-7能增加A549细胞MMP-7-mRNA的表达，LIPO-MAON可降低其表达（P〈0．05）。结论 非小细胞肺癌患者MMP-7表达阳性率为82．5％，MMP-7反义寡核苷酸能抑制肺腺癌细胞的增殖及MMP-7-mRNA表达，对正常血管内皮细胞增殖无抑制作用。MMP-7能促进肺腺癌细胞增殖。
