摘    要:目的:对含PTD-SOD重组质粒的工程菌进行诱导表达。方法:通过SOD活性和SDS-PAGE电泳分析,考察了诱导时间和诱导剂浓度对PTD-SOD表达量的影响。同时考察了诱导温度对产物表达形式的影响。结果:表达条件为IPTG浓度0.5mM,诱导时间4h,诱导温度20℃。结论:在上述条件下重组SOD在摇瓶中的表达效果最好。

Abstract:Objective:To study the induction and expression of strains with recombinant plasmid of PTD-SOD.Methods: The effects of the concentration of inducer as well as the inducing time on the expression amount of PTD-SOD were investigated by SDS-PAGE analysis.The influence of induction temperature on the style of expressed PTD-SOD was also analyzed.Results:The expression conditions were as follows: IPTG concentration 0.5mM,induction temperature 20℃ and induction time 4 hours.Conclusion: The above conditions are the optimum expression conditions for recombinant strain flask cultivation.
