TGF-β诱导骺板肥大区软骨细胞向成骨样细胞分化

目的　通过研究转化生长因子 - β(TGF- β)对骺板软骨细胞分化的作用 ,揭示 TGF- β软骨内成骨的作用机制。方法　采用鸡胚股骨无血清培养、酶组化检测碱性磷酸酶 (AL P)、免疫组化检测 型胶原、纤维粘连蛋白 (FN)等方法 ,观察 TGF- β在不同作用时间和剂量时对骺板肥大区软骨细胞分化的作用。结果　 TGF- β可使肥大区软骨细胞表达 型胶原和 FN,使 AL P表达增加 ,并存在着量效和时效关系。结论　 TGF- β可以诱导鸡胚股骨骺板肥大区软骨细胞向成骨样细胞分化 ,与 TGF- β的作用剂量和时间密切相关。

Transforming Growth Factor-beta Can Induce Differentiation of Hypertrophic Chondrocytes into Osteoblast-like Cells in Epiphyseal Plate

Objective　To investigate the effect of transforming growth factor-beta (TGF-β) on the differentiation of hypertrophic chondrocytes in epiphyseal plate in order to understand its role in the mechanism of bone and cartilage formation. Methods　We observed the effect of TGF-β in varied time and dosage on the differentiation of hypertrophic chondrocytes in embryonic chick femora which were cultured in serum-free medium, using histochemical staining to detect the expression of alkaline phosphatase (ALP) and immunohistochemical staining to detect the expression of type Ⅰcollagen and fibronectin. Results　TGF-β enabled the chondrocytes to express type Ⅰcollagen and fibronectin, and it enhanced the expression of ALP in hypertrophic zone. Time-effect and dosage-effect relationships were observed. Conclusion　TGF-β can induce the differentiation of hypertrophic chondrocytes into osteoblast-like cells in epiphyseal plate of embryonic chick femora. The inducing effect might be closely related to time and dosage of TGF-β.
