血管内皮细胞线粒体膜电位与缺氧及血管紧张素Ⅱ的相关性

摘    要:目的观察缺氧及血管紧张素Ⅱ(Ang-Ⅱ)对血管内皮细胞(VEC)线粒体膜电位的影响。方法建立VEC缺氧损伤模型;实验设为空白对照组、缺氧组、Ang-Ⅱ作用组、缺氧加Ang-Ⅱ作用组等四组。细胞经过Rhodamine 123负载后,用激光共聚焦显微镜测定VEC内Rhodamine 123的荧光强度代表线粒体膜电位。结果VEC分别经过缺氧及Ang-Ⅱ损伤后,线粒体膜电位明显降低(P<0.01);缺氧与Ang-Ⅱ联合作用可引起VEC线粒体膜电位较单纯缺氧或单纯Ang-Ⅱ作用进一步降低(P<0.05)。结论缺氧及Ang-Ⅱ可引起VEC线粒体膜电位明显降低,造成VEC损伤。

Abstract:Objective To study the effect of hypoxia and angiotesinⅡ (AngⅡ) on mitochondria membrane potential (MMP) of vascular endothelial cells (VECs). Methods Hypoxia-induced injury model of VECs was established, and then cultured VECs were divided into four groups: control group, simple hypoxia group, simple AngⅡ group, hypoxia+AngⅡ group, which were loaded with Rhodamine 123. MMP was detected by laser confocal microscopy to see if there were any differences among the groups. Results Compared with control group, decrease in MMP was observed either in the hypoxia group or in the Ang Ⅱ group (P<0.01). This change was clearer in the hypoxia+ AngⅡ group than in any of the other groups (P<0.05). Conclusion Hypoxia or AngⅡ could significantly decrease MMP of VECs, and injure VECs.
