头孢克肟在呼吸系统感染病人体内的药物动力学研究

 本实验对头孢克肟在１０例呼吸系统感染病人体内的药物动力学特性进行了研究。头孢克肟血浓度采用ＨＰＬＣ测定。ｐｏ头孢克肟２００ｍｇ后，在病人体内的药物配置为一房室一级吸收模型。消除半衰期（Ｔ＿（１／２Ｋｅ））为３．０９±０．０９ｈ吸收半衰期（Ｔ＿（１／２Ｋａ））为１．４４±０．２１ｈ，达峰时间（Ｔ＿（ｐＫ））为３．７０±０．２７ｈ，峰浓度（ｃ＿（ｍａｘ））为２．４４±０．２５μｇ／ｍｌ，血清总清除率（Ｃｌ＿ｓ）为０．５４±０．１３ｍｌ／（ｍｉｎ·ｋｇ），表现分布容积（Ｖ）为０．１５±０．０３Ｌ／ｋｇ，滞后时间（ｌａｇｔｉｍｅ）为０．７４±０．０８ｈ。

Pharmacokinetics of cefixime in patients with respiratory tract infec-tions

Cefixime（CFX） is a third generation cephalosporin,it can be orally administered.The pharmacokinetics of CFX was studied in ten patients with respiratory tract infections,Ofthese patients were 6 males and 4 females.The diagnosis among them was:asthmatiform bronchi-tis 5 cases,pulmonary infections 4 cases,upperrespiratory tract infections 1 case.Hepatic and re-nal functions were normal in 10 patients.Each patient was orally given 200 mg cefixime capsulewith 240 ml water in fast in morning,and drank water three time 240 ml each time at 2,4,6 h andhad a lunch at 4 h after administration,Venous blood samples were collected into tubes at 0,0.5,1.0,2.0,3.0,4.0,5.0,6.0,8. 0 and 12.0h after administration.Serum CFX Ievels was detectedby high-pressure liquid chromatography.The one compartment model was used.The mean peakserum level was 2.64±0.29 μg/ml.The mean peak time was 4.0h.Pharmacokinetic parametersof CFX in patients,plasma half-life of elimination(T1/2Ke),elimination rate constant（Ke）,half-lifeof absorption(T1/2Ka),absorption rate constant（Ka）,volume of appearance distribution（Vd）,serum clearance（Cl）and area under curve（AUC）were 3.09±0.09 h,0.22±0.01 h,1.44±0.21h,0.49±0.07h,0.15±0.03 L/kg,0.54±0.13 ml/（min·kg）,and 21.24±2.93 μg·h/ml,respec-tively.
