| 摘 要： | 目的 观察人肝硬化肝癌组织中丝裂原活化蛋白激酶家族两种主要亚型细胞外信号调节蛋白激酶(ERK)、c-Jun氨基末端蛋白激酶(JNK)的表达.方法 应用半定量逆转录-聚合酶链反应(RT-PCR)方法检测10例人中晚期肝硬化肝癌及其癌旁正常组织中ERK、JNK mRNA的相对表达量,并采用Western blot检测上述组织中ERK、JNK蛋白产物表达.结果 90%(9/10)的肝癌组织中ERK、JNK mRNA和蛋白表达增强,而癌旁组织中80%(8/10)该基因表达缺失和蛋白表达减少,两者差异有统计学意义(P<0.05).结论 肝硬化肝癌组织中ERK、JNK表达处于活化状态,可能在肝硬化肝癌的发生、发展过程中发挥重要作用.

| Abstract: | Objective To observe the expression of-ERK and JNK in human hepatic cirrhosis carcinoma (hHCC).Methods The samples were obtained from 10 cases of hHCC.Semi-quantitative RT-PCR and Western blot were used to detect the expression of ERK,and JNK mRNA and protein in 10 samples and adjacent noncanerous tissues.Results The expression of ERK, and JNK mRNA and protein was detected in 90% of hHCC and 20% of adjacent noncanerous tissues (P < 0.05).Conclusion ERK, and JNK activation in hHCC may play some roles in the pathogenesis of HCC. |
