针刺对高脂大鼠主动脉脂联素受体基因表达的影响

摘    要:目的探讨AdipoR的不同亚型在高脂饮食机体主动脉表达的变化以及针刺对该变化的影响。方法将高脂喂养12周的SD大鼠检测血脂、一氧化氮(NO)和髓过氧化物酶(MPO),根据体重随机分为模型和针刺组,干预4周后检测血脂、NO、MPO、主动脉AdipoR1和AdipoR2基因表达,并设普食对照组。结果干预前,与对照组比较,模型组和针刺组大鼠TG、TC、MPO明显升高,NO明显降低。干预后,模型组TG、TC、MPO、NO无明显变化,主动脉AdipoR1和AdipoR2基因表达明显降低;针刺后TG、TC和MPO明显降低,NO明显回升,主动脉AdipoR2基因表达明显上调,AdipoR1基因表达无明显变化。结论高脂饮食造成机体主动脉AdipoR1和AdipoR2基因表达低下,影响了脂联素信号对主动脉的保护作用;针刺能够上调脂联素信号保护高脂对主动脉的损伤,其作用主要通过AdipoR2介导的。

Abstract:Objective To study the effect of acupuncture on adiponectin receptor(AdipoR)expressions of aorta in hyperlipidemia rats.Methods Rat models were established with high-fat feed for 12 week.All rats were randomly divided into model group and acupuncture group based on the body weight.The levels of serum lipid,nitric oxide(NO)and myeloperoxidase(MPO) were measured,and the aortal mRNA expressions of adipoR1 and adipoR2 were detected with real time RT-PCR after 4 weeks treatment.And rats with normal forage were as controls.Results Compared with the controls,serum levels of triglyceride(TG),total cholesterol(TC)and MPO were increased obviously and the NO content was decreased markedly in both model and acupuncture groups pre-treatment.after treatment for 4 weeks,there were no changes in the levels of serum TG,TC,MPO and NO.The aortal mRNA expressions of AdipoR1 and AdipoR2 were depressed obviously in model group.Compared to the model,the levels of serum TG,TC and MPO in acupuncture group were decreased significantly.Serum NO content and aortal AdipoR2 mRNA expression were increased markedly.Conclusion Depressed aortal mRNA expressions of AdipoR1 and AdipoR2 due to high-fat dietary could affect the protective function of adiponectin signal on aorta.Acupuncture could upregulate adiponectin signal and protect aorta from damage induced by high-fat dietary,which could be mediated primarily by AdipoR2.
