参元丹晚期药理预适应对大鼠缺血再灌注心肌PKC及HSP70的影响

摘    要:目的应用免疫组化法观察蛋白激酶C(PKC)及热休克蛋白(HSP70)表达,探讨参元丹(SYD)的晚期药理性预适应作用及其机制。方法选取清洁级健康成年Wistar大鼠60只。随机分为6组,分别为对照组、再灌注组、晚期假性缺血预处理组、晚期缺血预处理组、晚期药物预处理加缺血预处理组、晚期药物预处理组。每组10只。分别造模,取血、心肌组织。应用HE染色方法对组织切片进行染色,彩色病理图像分析心肌梗死面积。20倍镜下观察PKC及HSP70表达情况。结果彩色病理图像分析,SYD能够代替心绞痛缺血、缺氧刺激,减小心肌梗死面积,具有晚期药理性预适应的心脏保护作用。晚期假性缺血预处理组、晚期药物预处理加缺血预处理组、晚期药物预处理组无PKC的表达,提示PKC未参与晚期缺血预适应的细胞内信号转导。晚期缺血预处理组、晚期药物预处理加缺血预处理组均有HSP70大量表达,晚期药物预处理组亦有HSP70表达,提示晚期缺血预处理的心肌保护作用机制与HSP70有关,SYD具有独立的晚期药理预适应作用,晚期药物预处理组+缺血预处理组的HSP70表达有SYD作用存在;SYD的心肌保护作用机制与HSP70有关。结论SYD具有晚期药理预适应样心肌保护作用;SYD晚期预适应心肌保护作用的机制可能与HSP70的介导有关;SYD晚期预适应可能与缺血预适应协同发挥心肌保护作用。

Effects of Delayed Preconditioning with Shenyuandan Capsule on Protein Kinase C and Hot Shock Protein 70 in ischemia-reperfusion Myocardium in Rats

Abstract:Objective To study the effects of delayed preconditioning with Shenyuandan capsule on protein kinase C(PKC)and hot shock protein 70(HSP70)in ischemia-reperfusion myocardium in rats.Methods Sixty Wistar rats were randomly divided into six groups(n=40 each group):Control group,reperfusion group,delayed pseudo-ischemic preconditioning group,delayed ischemic preconditioning group,delayed drug pre-treatment with ischemic preconditioning group,and delayed drug pre-treatment group.Then the model was induced,blood samples and myocardium were collected.PKC and the HSP70 expression were observed.Results Colored pathology image analysis suggested that Shenyuandan capsule could block ischemia and oxygen deficit stimulation in angina pectoris,and could reduce the area of myocardial infarction.There were no expression of PKC in delayed pseudo-ischemic preconditioning group,delayed drug pretreatment with ischemic preconditioning group,and delayed drug pre-treatment group.There was a great number of HSP70 expression in delayed ischemic preconditioning group and delayeddrug pretreatment with ischemic preconditioning group,and delayeddrug pre-treatment group.Shenyuandan capsule had an independent effect in delayed pharmacological preconditioning.There was the role of Shenyuandan capsule in delayed drug pre-treatment with ischemic preconditioning group.Conclusion Shenyuandan capsule had delayed pharmacological preconditioning-like myocardial protective effect.It might be related to HSP70.Shenyuandan capsule delayed preconditioning and ischemic preconditioning might play an association role in the myocardial protection.
