泽兰对人冠状动脉平滑肌细胞增殖的影响

目的研究中药泽兰对人冠状动脉平滑肌细胞（HCASMC）增殖的影响。方法利用体外细胞实验,通过人碱性成纤维生长因子和人表皮生长因子刺激细胞增殖,随机分为空白组、不同浓度泽兰水提物（LLST）组（320μg/mL、640μg/mL、1 280μg/mL、2 560μg/mL、5120μg/mL）和不同浓度泽兰醇洗脱物（LLCX）组（40μg/mL、80μg/mL、160μg/mL、320μg/mL、640μg/mL）。采用MTT法和流式细胞术分别测定细胞OD值、G0/G1期、S期、G2/M期和apoptosis期细胞百分比,观察泽兰不同浓度药液对细胞增殖的影响。结果 MTT结果显示泽兰LLST和LLCX 10个剂量均可抑制细胞增殖（P〈0.05）。细胞周期结果显示：泽兰LLST 320μg/mL、640μg/mL、1 280μg/mL浓度组和泽兰LLCX40μg/mL、80μg/mL、160μg/mL浓度组的G0/G1期细胞百分比较空白组增高（P〈0.05）;S期细胞百分比较空白组降低（P〈0.05）。抑制增殖作用的最佳药物浓度分别为1 240μg/mL和160μg/mL。结论中药泽兰具有抑制HCASMC增殖的活性。

Influence of Herba Lycopi on Proliferation of Human Coronary Artery Smooth Muscle Cells

Objective To study the influence of herba lycopi on proliferation of human coronary artery smooth muscle cells（SMCs）.Methods HCA SMCs were stimulated by human basic fibroblast growth factor（hbFGF） and human epidermal growth factor（hEGF）.Then the stimulated cells were randomly divided into 11 groups：Blank group,and 5 groups with different concentrations of water extracts（LLST）（320,640,1280,2560,5120 μg/mL） and 5 groups with ethanol elution extracts（LLCX）（40,80,160,320,640 μg/mL） of herba lycopi.OD value,percentage of G0/G1 phase cells and S phase and G2/M phase in cultured cells were detected to observe the influence of different concentrations of LLST and LLCX on HCA SMCs proliferation.Results The ten doses of the LLST and LLCX of herba lycopi could inhibit HCA SMCs proliferation（P〈0.05）.The percentages of G0/G1 phase cell in LLST 320,640,1 280 μg/mL groups and LLCX 40,80,160 μg/mL groups were higher than that in blank group（P〈0.05）,and the percentage of S phase cells was lower than blank group（P〈0.05）.The best drug concentrations of inhibiting proliferation were 1240 μg/mL and 160 μg/mL.Conclusion Herba lycopi could inhibit the proliferation of HCA SMCs.
