芪红合剂含药血清对人外周血内皮祖细胞数量及其功能的影响

摘    要:目的观察芪红合剂含药血清对人外周血内皮祖细胞(EPCs)数量及其功能的影响。方法采用血清药理学方法,制备成芪红合剂低、中、高剂量含药血清及空白血清。用密度梯度离心法分离获得单个核细胞,培养7 d后,经激光共聚焦显微镜、流式细胞仪鉴定为正在分化的EPCs;收集贴壁细胞,分别用含低、中、高剂量芪红合剂含药血清及空白血清的条件培养液培养12 h、24h、48 h后,倒置相差显微镜下观察EPCs生长情况。分别采用细胞计数法、黏附能力测定实验观察EPCs的数量和黏附能力。结果与对照组比较,芪红合剂含药血清能增加EPCs的数量,且能改善EPCs黏附能力,并且其影响呈一定的剂量相关性、时间依赖性。低剂量芪红合剂组较中、高剂量组作用明显(P〈0.05),且在同一剂量组中,芪红合剂含药血清对EPCs数量和黏附功能的影响随时间推移逐渐增强,于72 h达到高峰作用(P〈0.05)。结论芪红合剂能促进体外培养人外周血EPCs的数量和黏附功能。

Abstract:Objective To investigate the effects of Qihong decotion drug-containing serum on numbers and function of endothelial progenitor cells(EPCs) derived from peripheral blood.Methods Using the method of serum pharmacology,the drug-containing serum(Qihong decoction low dosage group,medium dosage group,high dosage group)and control serum were prepared.The mononuclear cells were isolated from human peripheral blood by density gradient centrifugation.After 7d of culture,the laser scanning confocal microscope and the flow cytometry(FCM)was used to determine the EPCs of human peripheral blood.The cultured EPCs were incubated with different drug-containing and control serum cultivate liquid for different duration(24 h,48 h and 72 h).The activity of EPCs was observed under reverse microscope.Numbers of EPCs,and the ability of cell adhesion was assessed by cell counting,and adhesion assay.Results Compared with control group,the numbers and adhesion functions of EPCs were remarkably increased by Qihong decoction drug-containing serum,showing a time-effect and dose-effect relation.The Qihong decoction low dosage group had the most prominent effect(P〈0.05).For the same Qihong decoction dosage group,the effect to cell numbers and the ability of cell adhesion increased as time went by and reached peak at 72 h(P〈0.05).Conclusion Qihong decoction drug-containing serum can premote numbers and cell adhesion function of EPCs derived from peripheral blood.
