KISS-1蛋白和MMP-9在妊娠滋养细胞病变中的表达

目的:探讨KISS-1基因蛋白(metastin)和基质金属蛋白酶-9(MMP-9)在妊娠滋养细胞病变(妊娠滋养细胞疾病和自然流产)中的表达及二者的相关性。方法:采用免疫组化S-P法分别检测20例正常早孕绒毛、25例自然流产绒毛、30例完全性葡萄胎、15例侵袭性葡萄胎和15例绒毛膜癌中KISS-1蛋白和MMP-9的表达。结果:KISS-1蛋白在自然流产绒毛中的表达高于正常早孕绒毛(P<0.05);侵袭性葡萄胎和绒毛膜癌中的表达显著低于完全性葡萄胎(P均<0.01);在侵袭性葡萄胎和绒毛膜癌中的表达与临床分期呈负相关。MMP-9在自然流产绒毛中的表达低于正常早孕绒毛(P<0.05);在完全性葡萄胎中的表达高于正常早孕绒毛(P<0.05);侵袭性葡萄胎和绒毛膜癌中的表达高于完全性葡萄胎(P均<0.05);在侵袭性葡萄胎和绒毛膜癌中的表达与临床分期呈正相关。在自然流产绒毛、侵袭性葡萄胎和绒毛膜癌中KISS-1蛋白表达和MMP-9表达均呈负相关。结论:KISS-1蛋白和MMP-9表达的变化可能参与了自然流产和妊娠滋养细胞疾病(葡萄胎、侵袭性葡萄胎和绒毛膜癌)的发生、发展过程,在此过程中两种基因存在着相互作用。

Expression of KISS - 1 and MMP -9 in gestational trophoblastic lesions

Objective:To explore the expression and relationship of KISS-1 and MMP-9 in gestational trophoblastic lesions(gestational trophoblastic disease,GTD and hydropic spontaneous abortion).Methods:The expression of KISS-1 and MMP-9 at protein level in 20 cases normal first trimester placenta,25 cases of hydropic spontaneous abortion,30 cases of complete hydatidiform mole(CHM),15 cases of invasive mole and 15 cases of choriocarcinoma were comparatively studied with immunohistochemical staining method.Results:The expression of KISS-1 in hydropic spontaneous abortion was higher than that of normal first trimester placenta(P<0.05),the expression of KISS-l in invasive mole and choriocarcinoma were all significantly lower than that in CHM(P<0.01,P<0.01),and was negatively related to the clinical stage in invasive mole and choriocarcinoma.The expression of MMP-9 in hydropic spontaneous abortion was lower than that of normal first trimester placenta(P<0.05),in CHM was higher than that of normal first trimester placenta(P<0.05),in invasive mole and choriocarcinoma were all significantly higher than that in CHM(P<0.05,P<0.05),and was positively related to the clinical stage in invasive mole and choriocarcinoma.The negative relationship was found between the expression of KISS-1 and MMP-9 in hydropic spontaneous abortion,invasive mole and choriocarcinoma.Conclusion:The interaction of KISS-I and MMP-9 abnormalities might play roles in the initiation and progression of GTD and hydropic spontaneous abortion.
