摘    要:目的 构建铜-锌超氧化物歧化酶(SOD1)基因缺失的酿酒酵母菌株, 为进一步以酵母为模型开展家族性肌萎缩硬化症发病机制及防治研究奠定基础.方法 运用LFH-PCR构建了带有遗传霉素抗性基因(KanMX4)的SOD1基因中断框,转化酿酒酵母BY5677,以添加G418的平板筛选出SOD1基因缺失菌株,并经PCR鉴定.结果 PCR扩增证实SOD1基因已被成功敲除;与野生菌株相比,SOD1基因缺失菌株生长缓慢.结论 成功构建了SOD1基因缺失的酿酒酵母菌株.

Abstract:Objective To construct the SOD1 gene deletion mutant of Saccharomyces cerevisiae and lay a foundation for studying the pathogenesis and treatment of familial amyotrophic lateral sclerosis based on the yeast model.Methods The SOD1:KanMX4 gene deletion fragment was constructed by long flanking homology LFH-PCR and transferred to yeast strain BY5677.The SOD1 gene deletion mutant was selected on plates containing G418 and confirmed by PCR.Results The PCR analysis showed that SOD1 gene in the yeast strain BY5677...
