银杏叶联合依那普利对单侧输尿管梗阻肾间质纤维化的影响

摘    要:目的观察银杏叶联合依那普利对肾间质纤维化的防治作用及其效果。方法将75只雄性SD大鼠随机分为5组,即假手术组、模型组、依那普利组、银杏叶组及依那普利加银杏叶组,每组15只。除假手术组外,采用单侧输尿管梗阻(UUO)建立肾间质纤维化模型。银杏叶组、依那普利组、依那普利组加银杏叶组分别给予银杏叶300mg/(kg·d)、依那普利粉末10mg/(kg·d)及银杏叶加依那普利联合灌胃,每日1次,假手术组及模型组给予等容积0.9%氯化钠注射液灌胃治疗。于实验第3、7、14d分批处死大鼠,各组每次处死5只。留取大鼠结扎侧肾组织进行形态学和免疫组织化学观察。观察指标:肾组织经HE染色观察梗阻侧肾组织病理学的改变,用免疫组织化学SP方法检测肾组织中转化生长因子β1(TGF—β1、)、核因子KB(NF—KB)、仪平滑肌肌动蛋白(α-SMA)的表达情况及肾组织中胶原蛋白与肾质量之比。结果与假手术组相比,模型组中肾间质水肿,单个核细胞浸润,肾间质纤维组织相对面积显著扩大,TGF—β1、NF—κB、α-SMA的表达及肾组织中胶原蛋白含量表达明显上调(P〈0.05)。药物干预后,上述上调指标均被显著抑制(P〈0.05),其中依那普利组加银杏叶组与依那普利组、银杏叶组比较差异有统计学意义(P〈0.05),而依那普利组与银杏叶组比较差异无统计学意义(P〉0.05)。结论银杏叶片能够减少UUO大鼠肾间质炎性细胞浸润,减少肾间质胶原蛋白的含量,有防止UUO大鼠肾间质纤维化的作用,银杏叶和依那普利两者合用抗RIF有协同作用。

Effects of Ginkgo biloba tablet combination enalapril in renal interstitial fibrosis rats with unilateral ureteral obstruction

Abstract:Objective To observe the preventive and therapeutic effects of Ginkgo biloba tablet combinaton enalapril in renal interstitial fibrosis rats. Methods Unilateral Ureteral Obstruction (UUO) model was induced by ligating the left ureter in rats. 75 male sprague dawley rats were randomly divided into 5 groups : sham operation group, model group, control groups( enalapril group, ginkgo biloba tablet group, ginkgo bitoba tablet combination enalapril group). Every group has 15 rats. Rats in treated group were differently injected ginkgo biloba tablet (300 mg/ (kg · d) or enalapril( 10 mg/( kg · d)or ginkgo biloba tablet combination enalapril. HE,The rats in sham and control groups were injected the same volume of normal water. Such were observed at 3th ,7th and 14th day:pathological changes of the obstructed kidney and the changes of TGF - β1, NF - κB, α - SMA and nephridial tissue collagen protein/kidney weight. Results Massive inflammatory infiltrates and collagen expression in renal interstitial in the mod- el group were observed . Compared with the sham operation group, percentages of area of TGF - β1, NF - κB and α - SMA expressions in the model group were markedly increased ( P 〈 0. 05). After intervention with treated group, the upregulation of the above - mentioned parameters were all significantly inhibited ( P 〈 0.05 ). There was significant difference of intervention effects between ginkgo biloba tablet combination enalapril group and other - treated group ( P 〈 0.05 ). There was no significant difference of intervention effects between enalapril group and ginkgo biloba tablet group ( P 〉 0.05 ). Conclusion Ginkgo biloba tablet group can reduce inflammatory infiltrates, renal interstitium collagen protein content, and the degree of interstitial fibrosis. Ginkgo biloba tablet combination enalapril could significantly attenuates renal interstitial fibrosis.
