乳酸杆菌胃分离株WR22体外抑制幽门螺杆菌黏附胃上皮细胞研究

摘    要:目的研究乳酸杆菌WR22体外对胃上皮细胞的黏附能力和抑制幽门螺杆菌黏附上皮细胞的效果。方法乳酸杆菌WR22在体外与胃上皮细胞爬片共同孵育2h后,采用光镜、电镜技术观察其黏附效果。WR22与幽门螺杆菌SS1对胃上皮细胞竞争性黏附2h后,荧光抗体染色法定性评价黏附于胃上皮细胞上的幽门螺杆菌密度。结果乳酸杆菌WR22在活菌和死菌状态下都能大量地黏附于上皮细胞,显著降低幽门螺杆在胃上皮细胞上的黏附密度,但死菌状态下抑制能力次于活菌状态。结论乳酸杆菌胃分离株WR22具有显著抑制幽门螺杆菌体外黏附胃上皮细胞的能力,有待进行体内效果评价。

Abstract:To investigate the adherence ability of Lactobacillus salivarius strain WR22 to gastric epithelial cell SGC-7901 and its effect of inhibiting the adherence of Helicobacter pylori to the cells, the light microscope and scanning electron microscope were used to detect L, salivarius stain WR22 adhered to SGC-7901 cells after co-culturing for 2 hours, By using FITC-conjuncted antibody ,the adhered Helicobacter pylori density was evaluated under fluorescence microscope after pre-treatment with live or nonviable WR22. It was found that both the living and nonviable L, salivarius stain WR22 could adhere to SGC-7901 tightly and obviously inhibited the adherence of Helicobacter pylori, but living WR22 has better inhibiting ability than the nonviable state. It is concluded that L. salivarius stain WR22, originally isolated from human stomach, can inhibit the adherence of Helicobacter pylori to gastric epithelial ceil SGC-7901 in vitro.
