青海省67份轮状病毒结构蛋白VP7基因分型和分布情况

摘    要:目的 研究2016-2017年青海地区腹泻患者轮状病毒基因分型及流行病学分布。方法 收集门诊及住院部腹泻的粪便标本238份,采用实时荧光PCR法对轮状病毒A组进行检测,阳性标本进行VP7基因扩增和测序。结果 238份粪便标本中,通过实时荧光PCR 检测到轮状病毒A组阳性67份,阳性率为28.15%(67/238);对67份轮状病毒A阳性标本进行VP7蛋白检测和测序,测序后得到29份核苷酸序列,用Clustral X Bootstap NJ Tree软件构件进化树,分析发现2016年3月-2017年12月青海轮状病毒以G9P8型为主,共26株,占89.66% (26/29),G2P4型2株(2/28),G3P8型1株(1/28), 轮状病毒腹泻发病高发季节为9-12月,其中以12月份检出最高,占总数的61.19%(41/67)。病人以成人为主,成人和5岁以下儿童比例为1.73DK]∶1。结论 2016-2017年青海地区轮状病毒以流行病毒株G9P8型为主。

Abstract:Objective To study the genotype and epidemiology of rotavirus in diarrhoea patients in Qinghai from 2016 to 2017. Methods 238 specimens of diarrhea in the outpatient and inpatient departments were collected. The rotavirus group A was detected by real-time fluorescent PCR, and the positive specimens were amplified and sequenced by VP7 gene. Results In 238 stool samples, 67 positive samples from rotavirus group A were detected by real-time fluorescent PCR, accounting for 28.15% (67/238). VP7 protein was detected and sequenced in 67 rotavirus positive samples. 29 nucleotide sequences were obtained, and the evolutionary tree of Clustral X (Bootstap NJ Tree) software was used. It was found that from March 2016 to December 2017, Qinghai rotavirus was mainly G9P8 type, a total of 26 strains, accounting for 89.66% (26/29), 2 strains of G2P4 (2/28) and 1 strain of G3P8 (1/28). The high incidence of rotavirus diarrhea was from September to December, with the highest detected in December, accounting for 61.19% (41/67) of the total. The majority of patients were adults, and the ratio of adults to children under 5 was 1.73DK]∶1. Conclusion In 2016-2017, the rotavirus in Qinghai is dominated by the epidemic strain G9P8. However, in 2012-2015, the mixed infection of Qinghai G3P8 and G9P8 indicates that the epidemic strains in Qinghai are constantly changing.
