乳腺癌中核转录因子-κB p50表达及其与细胞凋亡的关系

摘    要: 目的 研究核转录因子-κB p50 (NF-κB p50)在乳腺癌组织中的表达和意义及其与细胞凋亡的关系。方法 应用Elivision免疫组化染色法和脱氧脱氧核糖核酸末端转移酶介导的缺口末端标记(TUNEL)技术,对65例乳腺癌组织中NF-κB p50、凋亡细胞表达进行原位观察和比较,并结合临床资料进行分析。结果 65例乳腺癌标本NF-κB p50表达阳性率为70.8 %(46/65),细胞凋亡指数(AI)为(4.59±1.94)%; NF-κB p50表达强度与患者的肿瘤的大小、组织分级、孕激素受体(PR)无关(P>0.05);与腋淋巴结转移数、雌激素受体(ER)状况明显相关(P<0.05)。NF-κB p50表达阴性的乳腺癌中AI明显高于NF-κB p50 表达阳性组(P<0.05)。NF-κB p50活性强度与AI呈明显的负相关(P<0.001)。结论 NF-κB p50的异常激活在早期乳腺癌的发生和转化中起重要作用,其抑制凋亡活性与肿瘤侵袭转移及恶性潜能密切相关,对NF-κB p50活性及细胞凋亡的检测有助于乳腺癌的临床分析,并有望成为预后的观察指标。

Abstract:Objective To study the expression of NF-κB p50 and its significance in breast cancer(BC), and to inquire into the relation between NF-κB p50 and apoptosis. Methods The expression of NF-κB p50 and apoptosis were detected by observing the immunohistochemical dyeing and in situ terminal deoxynu-cleotidyl-transferase mediated dUTP-biotin nick ending labeling (TUNEL) assay of 65 specimens of BC. Re-sults The positive expression ratio of NF-κB p50 was 70.8 %(46/65), and the apoptotic index (AI) of BC was(4.59±1.94) %. The expression of NF-κB p50 in BC was not related to tumor size, histological grade and progesterone receptor (PR)(P > 0.05), but to the lymphnode metastasis of axillary fossa, estrogen receptor (ER)(P < 0.05). AI in NF-κB p50 positive group was higher than NF-κB p50 negative group(P < 0.05). The intensity of NF-κB p50 expression was negatively correlated with apoptosis of BC(P < 0.001). Conclu-sions The abnormal activation of NF-κB p50 and cellular apoptosis may take an important role in the occur-rence, transformation of the primal BC and the activity of inhibit cell apoptosis was highly correlated with metastasis and malignant potential of BC. The detection of NF-κB p50 expression and apoptosis are helpful in clinical analysis, and may be a parameter in prognosis in BC.
